Abstract
Background : Paracetamol and tramadol is an example combination of analgesic drugs which is freely used. Studies have confirmed that paracetamol plus tramadol offers improved efficacy compared with either agent alone, and with no increase in the severity of adverse effects. However, the long term usage of analgesic may lead to an increased risks of renal dysfunction. Aim : To know the effect of analgesic combination of paracetamol and tramadol toward serum urea levels of wistar rats. Methods : Experimental study Post-Test Only Control Group Design. The samples were 28 male wistar rats, randomized into 3 groups. Group I was not given paracetamol and tramadol (control), Group II was given paracetamol 9 mg, Group III was given tramadol 0,9 mg, and Group IV was given the combination of paracetamol 9 mg and tramadol 0,9 mg. Administration through gastric instillation thrice a day for 14 days. Blood samples were collected at the 15th day through retroorbital vascular and the urea leves were measured using standard methods. The data was analyzed using One-Way ANOVA Test and Post-Hock Test. Results : There was no significant increase in the average value of the urea levels from the control group toward Group I, Group II, and Group III. One-Way ANOVA test showed that there was no significant difference (p=0,81) among all groups. Post-Hock test also showed that there were no significant differences in the control group toward Group I (p=0526), control group toward Group II (p=0,36), control group toward Group III (p=0,25), Group I toward Group II (p=0,77), Group I toward group III (p=0,88), and Group II toward Group III (p=0,66). Conclusion There is no significant increase in serum urea levels between administration of paracetamol and tramadol combination and control group Keywords: Analgesic, paracetamol, tramadol, urea levels.
