Abstract

Background: The incidence and mortality of breast cancer in Indonesia is high but the outcome of standard therapy for breast cancer is still not so good. Nigella sativa often used as an alternative in medicine treating cancer, including breast cancer. Thymoquinone is the highest component (57,87%) of volatile oil in Nigella sativa can inhibit the development of cancer cell and has cytotoxic effect. The objective of this study is to prove the difference of mitotic index and diameter alteration of tumor mass and to analyze the correlation between mitotic index and diameter alteration of tumor mass. Methods: An in vivo experimental study on breast adenocarcinoma C3H mice with post test only control group design (mitotic index) and pre and post test control group design (diameter alteration of tumor mass). There were 24 mice, divided into 4 groups (random). Three treatment groups were administered Nigella sativa extract in levelled doses of 0.035, 0.0715, 0.14 mg/day for 3 weeks, and one group were act as control. The diameter alteration of tumor mass and mitotic index were analyzed with ANOVA test and the corerelation was analyzed with Pearson’s correlation test. Results: There was significant difference in mitotic index and diameter alteration of tumor mass reduction in treatment groups compared with the control group (p<0,001). There was a significant positive correlation between mitotic index and development/diameter alteration of tumor mass (p<0.001, r=0.910). Conclusion: Nigella sativa is able to decrease mitotic index and reduce tumor mass progrssion. Key words: Nigella sativa, mitotic index, diameter of tumor mass, breast adenocarcinoma
