Abstract
Latar belakang: Hasil Riskesdas tahun 2013 prevalensi hipertensi di Provinsi Jambi sebesar 24,6%. Tahun 2016 hipertensi primer di Kabupaten Sarolangun menempati posisi kedua setelah ISPA sebesar 25,3%. Kebiasaan masyarakat Melayu menggunakan daun alpukat (Persea americana Mill) sebagai antihipertensi daripada obat modern perlu dibuktikan melalui penelitian tentang perbedaan efektifitas antihipertensi obat modern Furosemide, ekstrak daun P. americana Mill dan sediaan nanopartikelnya secara in vivo untuk menurunkan Tekanan Darah Sistol (TDS) dan Tekanan Darah Distol (TDD) dan meningkatkan volume urin. Tujuan:Penelitian ini untuk membuktikan perbedaan efektifitas obat modern Furosemide, ekstrak daun P. americanaMill dan sediaan nanopartikelnya secara in vivo sebagai antihipertensimelalui aktivitas antihipertensi. Metode: Jenis penelitian yang digunakan adalah eksperimental dengan rancangan post-test dengan 6 kelompok perlakuan yaitu kontrol normal, kontrol positif furosemide, kontrol negatif induksi NaCl 16%, ekstrak, nanopartikel ekstrak, dan nanopartikel kitosan dengan dosis 100 mg/kg berat badan. Jumlah sampel adalah 24 ekor tikus putih jantan Wistar. Hasil: Uji one-way anova menunjukkansignifikan (p=0,000) antara kelompok TDS, uji pos hoc Tamhane’s menunjukkan nanopartikel ekstrak berukuran 875 nm lebih efektif menurunkan TDS dan TDD dari pada Furosemide, sediaan ekstrak dan nanopartikel kitosan. Uji Kruskal-Wallis menunjukkan signifikan antara kelompok TDD (p=0,03), dan peningkatan volume urin (p=0,02). Nilai Indeks Aktivitas Diuretik (IAD) menunjukkan nanopartikel ekstrak memiliki aktivitas diuretik tertinggi, sementara nanopartikel kitosan berukuran 73.8 nm lebih spesifik sebagai ACE inhibitor. Simpulan:Ekstrak daun P.americana Mill, sediaan nanopartikelnya dan nanopartikel kitosan lebih efektif menurunkan TDS dan TDD dibandingkan Furosemide, nanopartikel ekstrak merupakan sediaan paling efektif sebagai antihipertensi. Background: Basic health research’s result in 2013 the prevalence of hypertension in Jambi Province was 24.6%. Primary hypertension in Sarolangun Regency 2016 was ranked second after URI 25.3%. Malay people habit in Sarolangun Regency using traditional medicines like decoction of Avocado leaf (Persea americana Mill) to cure hypertension requires processing ingredient development with other method to obtain more effective efficacy of drug as nanoparticles synthesized from avocado leaf extract Objective: This study aimed to determine the potential of antihypertensive of P.americana Mill leaf extract and its nanoparticle in vivo Method: This study used experimental type with post-test design with normal control, positive control furosemide, negative control induction of 16% NaCl and 3 treatments, extract, nanoparticles extract, and chitosan nanoparticles. Dose given in treatment is 100 mg/kg BW. Total sample tested was 24 male white mouse Wistar Results: The results of One-way ANOVA test showed there was significant difference (p= 0.01) between Systolic Blood Pressure group (SBP) with pos hoc Tamhane’s test showed nanoparticles extract has measurement of 875 nm could reduce the highest average SBP. Kruskal-Wallis Nonparametric Test showed there was significant difference between SBP group (p=0.03), urine volume increased (p= 0.02). Diuretic Aactivity Index value showed nanoparticles extract has the highest diuretic activity compared to Furosemide and extracts as much as 2.2 Conclusion: Persea americana Mill leaf extract and its nanoparticle in vivo has the potential to reduce SBP, TDD, and gives diuretic effect in each group, where nanoparticles extract has strong potential as antihypertensive.
