Abstract

Background : Melatonin was a free radical used frequently as an antioxidant. Melatonin was one of medicine developed as a sepsis therapy. There was no severe toxicity even though high dose was given. Sepsis caused increased levels of lactic acid through anaerobic glycolysis that caused by tissue hypoxia. Melatonin was effectively used to lower lactic acid levels so as to prevent the occurrence of death. Objective : to find out melatonin’s influence to the level of lactate acid toward wistar rat in sepsis model and to obtain the information of melatonin in decreasing the level of lactate acid. Method : This research was an experimental research with a randomized control group using pre and post test design. The samples were 12 male wistar rats with certain criteria divided into 2 groups. The first group was given an intraperitoneal injection of lipopolysaccharide (LPS) and was not given melatonin as control group. The second group was given an intraperitoneal injection of lipopolysaccharide (LPS) and was given melatonin via oral sonde as experimental group. After a week, in the eighth day, the blood of each rat was taken from the retro-orbital blood vessel. The statistical test used were paired t-test, independent t-test, and Mann Whitney test. Results : In the independent test, the average score of the level of lactate acid from control group was higher than the experimental group. In the paired t-test, there was no significant difference between control and experimental group except in the post 1 – post 2 and pre-post 2 which had significant difference in control group. In the Mann Whitney test, there was also no significant difference in pre-post 1 and pre-post 2. Conclusions : Melatonin treatment did not cause significant decrease of the level of lactate acid.
