Abstract
Background: This study is aimed to determine the effect of stratified doses of vitamin C needed to achieve optimal phagocytic activity of macrophages from septic mice . Methods: This study is an animal experimantal with randomized post test only control group design. The samples were 20 septic Balb/C mice divided into 4 treatment groups: control(K); administered with 0,52 mg intravenous vitamin C/day(P1), 1,04 mg vitamin C(P2), 2,6 mg vitamin C(P3). All groups were treated for 3 days, phagocytosis ability of macrophages and the level of intraperitoneal vitamin C were measured, and analyze their relationship. ANOVA with Bonferroni Test was used to analysis. Spearman’s Correlation test was performed to analyze the correlation. Results: Within group test showed significant difference in macrophage phagocytosis ability (p<0.001). Between group test showed: K-P1 (p<0,001), K-P2 (p<0,001), K-P3 (p<0,001), P1- P2 (p < 0,001), P1-P3 (p < 0,01). There were no significant difference between the P2-P3 (p = 0.48). Intraperitoneal vitamin C levels were significantly different between K-P2(p<0,001), K-P3(p<0,001), P1-P2(p=0,003), P1-P3(p<0,001), P2-P3(p<0,001), but there were no significant different between the K-P1(p=0,131). Correlation test showed positive correlation between levels of intraperitoneal vitamin C and macrophage phagocytosis with correlation coefficient 0,58 (p<0,001). Conclusion: There is significant increase macrophages phagocytosis Balb/C mice with septic were given vitamin C, ideal doses vitamin C injection for septic condition is 1,04 mg/days and intraperitoneal vitamin C level was increased significantly according dose given. Levels of intraperitoneal vitamin C has positive correlation with macrophage phagocytosis activity. Keywords: Sepsis, vitamin C, macrophage phagocytosis, intraperitoneal vitamin C
