Abstract
Salah sate metabolit yang terakumulasi dalam medium karbohidrat melalui proses transformasi enzimatik dalam sikius asam trikarboksilat yaitu asam 2- hidroksipropana-1,2,3-trikarboksilat. Asam ini disekresikan oleh niikroorganisme akibat kegagalan sikius asam trikarboksilat sehingga terjadi disfimgsi akonitase dan isositrat dehidrogenase. Penelitian ini bertujuan untuk memperoleh asam 2- hidroksipropana-1,2,3-trikarboksilat melalui proses tninsibrmasi polisakarida dalarn medium Marano aniditiercearf menggunakan kapang A.spergillus Hider. Dalam proses pemisahannya didapatkan 12,193 % (w/w) amilum dan 11,965 % (w/w) residu kering. Residu ini mengandung 14,092 % (w/w) selulosa dan 49 % (w/w) amilum. Kedua bentuk tersebut kemudian digunakan sebagai medium dalam proses transformasi. Dalam penelitian ini dilakukan variasi pH antara 2 sampai 7 selama 8 hari. Dad herbagai variasi pH ini diketahui akumulasi asam 2- hidroksipropana-1,2,3-trikarboksilat terbesar dalam medium amilum pada pH ---- 5 hari ke-7 sebesar 12,0423 g/L dan pada medium residu Maranta arudinaceae pada pH = 3 hari ke-6 yaitu 2,9272 g/L. Pada proses kristalisasi didapatkan kristal asam 2-hidroksipropana-1,2,3-trikarboksilat dengan titik leleh 154,33 °C. One of the metabolict that accumulated in carbohydrate medium with enzimatic transformation process in tricarboxylic acid cycle is 2-hidroxypropane¬1,2,3-trikarboxylic acid. This acid secreted by microorganism because the fail of tricarboxylic acid cycle that causes aconitase and isocitric dehydrogenase: disfungtion. This research was purposed to get 2-hidroxypropane-1,2,3- tricarboxylic acid with polysaccaride transformation process in Maranta arudinaceae medium using Aspergillu,s. niger. I got 11.965 % (w/w) dry residue and 12.193 % (w/w) amilum from Maranta arudinaceae separation process. This dry residue contains 14.092 % (w/w) cellulose and 49 % (w/w) amilum. That two form has used as medium in the transformation process. In this research done pH variation between 2 to 7 with transformation time 8 days. From such various pI-I has known that the highest concentration oft-hidroxyproOane-1,2,3-trikarboxylic acid that accumulated in amilum medium was in pH = 5 for 7 days long with 12.0423 g/L and in the medium Maranta arudinaceae residue was in pH = 3 for 6 days long with 2.9272 g/I,,. I got 2-hidroxypropane-1,2,3-trikarboxylic acid crystal with melting point 154.33 °C from the crystaiitation process.
