Abstract

Genestein (4',5,7 trihidroksiisoflavon) merupakan turunan senyawa isoflavon yang banyak ditemukan dalam famili leguminosae. Senyawa isoflavon diketahui memiliki aktifitas antioksidan dan estrogenik. Senyawa isoflavon dari kedelai dipisahkan dengan ekstrasi bertahap dengan beberapa pelarut, dilanjutkan .krdmatogy. ail lapis tipis dengan fasa diam silika gel GF 254 dan larutan pengembang isobutanol : asam asetat glasial : air dengan perbandingan 3:1:1. \Varna noda dapat dilihat dengan sinar UV 365 tun untuk mengetahui senyawa isoflavon. Pemisahan genestein dari isoflavon digunakan kromatogyafi lapis tipis dengan larutan pengembang n butanol : asam asetat : air dengan perbandingan 6:1:3. Noda yang sesuai dengan RF standar genestein selanjutnya diekstrasi dengan metanol dan direkristalisasi _ Identifikasi hasil isolasi berdasarkan warn, bentuk dan titik leleh. Uji kelarutan has dikerjakan terhadap larutan NaOH, air dan FeC13. Hasil identifikasi menunjukkan kristal berwarna kuning, bentuk lembaran batang tipis dan titik leleh 257°C. Kristal larut claim larutan NaOH dan air_ Dalam larutan FeCI3 menunjukkan perubahan warns coklat merah keunguan. Genestein (4',5,7 trihidroxyisoflavone) is the isotlavone compound which can be found in leguminosae's family. The isoflavone compounds were known to responsible an antioxidant and estrogenic. The isoflavon compounds in soybean was separated by steped - solvent extraction and using by thin layer chromatography as stationary phase and developing solvent is isobutanol : glacial acetic acid : water (3:1:1). The spot was shown by UV irradian at 365 nm to known the isoflavone compounds.. The separation of genestein from isoflavon was done by thin layer chromatography using silica TLC plated and developing solvent is n butanol : acetic acid : water (6:1:3). The spot has the same RF with standard of genestein is purified by methanol and then recrystalized. The isolated compound was identified based on the form, colour and it's melting point. Identification also be done by determination of solubility of crystal in NaOH, water and FeC13 solution. From this research was got yellow, sheet and melting point 257°C. The crystal can be dissolved in NaOH, water and has reddish violet in FeC13.
