Abstract

ABSTRACT Background: Macrophage eliminates intracellular microorganism by secreting microbicidal agent nitric oxide and ingest bound microorganism into vesicle to be destroyed. Macrophage functions influenced by antioxidant. Roselle tea component has an antioxidant activity that can modulate macrophage functions. Objective: This study aimed to analyze the effect of roselle tea on macrophage nitric oxide production and macrophage phagocytosis capability. Method: A post test-only controlled group design experiment was carried out on 4-6 weeks old, male Balb/c mice, infected with Salmonella typhimurium 105 CFU. The treatments given in 7 days were giving orally with roselle tea 2x0,24 ml each day (P1), 2x0,5 ml each day (P2), 2x0,98 ml each day (P3). Positive control group infected with Salmonella typhimurium 105 CFU, received water 2x1 ml each day. Negative control group received water 2x1 ml each day. The parameters of macrophage functions were nitric oxide production, and macrophage phagocytosis capability represent on phagocytosis index. Nitric oxide production was measured by determine the NO level from macrophage supernatant using modified Griess method. Macrophage phagocytosis capability was measured by using latex bead particle which represented by phagocytosis index. Data were analyzed by Kruskal Wallis test. Result: There was no difference on the nitric oxide production between the experiment group and control group (p=0,053).There was significant decrease on the macrophage phagocytosis capability represent by phagocytosis index (p=0,001) between the experiment and positive control groups. Conclusion: Roselle tea does not interference the macrophages nitric oxide production, but decrease the macrophage’s phagocytic function.
