Abstract
Microalgae was source of carotenoid production especially Chlorophyta such as Dunaliella salina and Chlorella pyrenoidosa. D. salina from sea water produce 7 mg total carotenoid consist of 3.5 mg lutein, 0,5 mg α-carotene, 0,6 mg β-carotene and 35 mg chlorophyll. Protoplast fusion between two microalgae enable to combine both advantages character of parents. the aim of the research in conducting molecular characterization and identification of fusant comparing with their parents and other Chlorophyta. Fusant characterization was performed by sequences homology analysis. Identification of fusant was done by molecular analysis using 18SrDNA. this research result revealed two fusant DSCPTD and DSCPTC which having 79% homology with D. salina and 80% with C. pyrenoidosa. Molecular identification of fusant DSCPTC was showed close relationship to Chlorella sp. BUM 1105 with 96% similarity index. Fusant DSCPTD showed 81% similarity with Uncultured Chlorophyta ADEsp 1-7 and Chlorophyceae sp. WSH-29. Fusant similarity index about 79-80% with both parents indicated that fusant gaining combination character of C. pyrenoidosa and D. salina
