Abstract

Background: Piper crocatum (red betel) is a plant widely known in Indonesia and has been used as medicinal plant. Red betel compounds include alkaloids, flavonoids, saponins, triterpenoids, and tannin. The extract of red betel leaf has immunomodulatory effects. Objective: To prove the effect of leaf extract of Piper crocatum with graded doses to the splenic lymphocyte proliferation of Balb/c mice infected with Salmonella typhimurium. Methods: Experimental laboratory research with post test only control group design. Sample of 25 mice balb/c that have met the inclusion and exclusion criteria, adapted for 7 days. Sample divided into 5 groups by simple random sampling. K1 group given the extract of Piper crocatum orally 10 mg/mouse/day, K2 are infected with Salmonella typhimurium intraperitoneally, P1 given extracts of Piper crocatum orally 10 mg/mouse/day, P2 given extracts of Piper crocatum orally 30 mg/mouse/day, P3 given extracts Piper crocatum orally 100 mg/mouse/day, and all treatment groups infected with Salmonella typhimurium intraperitoneally. The splenic lymphocyte proliferation with MTT Assay method examined on 15th day. Data described in the form of tables, figures and statistical analysis. Results: The highest average of splenic lymphocyte proliferation is P3 group, while the lowest is K1 group. The significant difference (p <0.005) was found in K2-P1, P2-K2, and K2-P3. The diffference of K1-K2, P1-P2, P1-P3 and P2-P3 was found insignificant. Conclusion: The leaf extract Piper crocatum multilevel dose for 14 days increases the splenic lymphocyte proliferation of balb/c mice infected with Salmonella typhimurium.
