Abstract

Background : Combination of paracetamol and tramadol is an ideal analgesic combination and effective to relieve moderate to severe pain. They act on different mechanism of action that synergistically giving a better potential in relieving pain. Beyond these advantages, these drugs also have potential side effects, one of which is on the kidney. Aim : To determine the influence of paracetamol and tramadol combination on serum creatinine level in Wistar rats. Methods : This is an experimental study using post test only control group design. The samples were 28 Wistar rats, randomized into 4 groups: control group, a group receiving paracetamol 9 mg, a group receiving tramadol 0,9 mg, and a group receiving paracetamol 9 mg and tramadol 0,9 mg combination, administered 3 times a day orally using gastric instillation for 14 days. At 15th day, blood is collected from retro orbital vessel to measure the serum creatinine levels. Statistical analysis was conducted using ANOVA Test and Post-Hoc Test. Results : Obtained from statistical analysis where there is no significant increase in serum creatinine levels in Wistar rats given paracetamol compared to the control group (p = 0,117), the group given tramadol compared to the control group (p =0,076), the group given paracetamol and tramadol combination compared to the control group (p = 0,143), the group given paracetamol compared to the group given tramadol (p = 0,816), the group given paracetamol compared to the group given paracetamol and tramadol combination (p = 0,907), and the group given tramadol compared to the group given paracetamol and tramadol combination (p = 0,727). Conclusion : There is no significant increase in serum creatinine levels between administration of paracetamol and tramadol combination and control group.
