Abstract

Chlorophyll is the pigment that can absorbs light, that’s electromagnetic radiation in the visible spectrum. Determination of chlorophyll content in plant tissue is done by extracting chlorophyll pigments with alcohol, acetone or methanol after that the extract will observed absorbance at a wavelength of 663 nm and 645 nm using a spectrophotometer. Spectrophotometer is one method of instrumental analysis using the basic interaction of energy and matter. Spectrophotometer can be used to determine the concentration of a solution through absorption intensity at a particular wavelength. In the study determination chlorophyll concentration using soursop leaf plant 10 grams extracted using 20 ml of 70% alcohol and filtered using filter paper of different types namely whattman 41 filter paper, smooth and coarse filter paper. The extraction can test the absorbance value at a wavelength of 663 nm and 645 nm using a spectrophotometer Genesys Spectonic 20 visible. Absorbance obtained is calculated using total chlorophyll formula: 20,2D645 + 8,02D663 (mg / L), chlorophyll a: 12,7D663 - 2,69D645 (mg / L), chlorophyll b: 22,9D645 - 4,68D663 (mg / L). By the calculation determining the concentration of chlorophyll with different filter paper obtained the highest scores in the concentration of chlorophyll b,total chlorophyll concentration values highest is the type of fitering with filter paper whattman 41 is equal to 6,132 mg/L.
