Abstract
β-carotene is a carotenoid that is most often found in higher plants and cause the roots of carrots orange. Determination of β-carotene levels in plant tissue is done by extracting β-carotene with non-polar solvents such as alcohol, acetone or methanol and then extract the observed absorbance at 452 nm wavelength using a spectrophotometer. Spectrophotometer is one method of instrumental analysis that uses basic interaction of energy and matter. Spectrophotometer can be used to determine the concentration of a solution through absorption intensity at a particular wavelength. In a study determining the concentration of chlorophyll is using carrot plants extracted using 100 ml of methanol and filtered using filter paper. The extraction can test the absorbance value at a wavelength of 452 nm using a spectrophotometer Genesys Spectonic 20 visible. Absorbance obtained is calculated using the formula of linear equations pure β-carotene: y = 0,131x + 0.189. From the calculation of the levels of β-carotene analysis with different concentrations of the highest value obtained at a concentration of 0.4 g / ml is equal to 1.73 ppm.
