Abstract
Backgroud. Incidence of Epidermoid skin cancer is high. Phaleria macrocarpa in previous research can stimulate immunity reponse. Objective. To know the response of supplementation of Phaleria Macrocarpa (PM) to Paclitaxel–cysplatin (Pacli-Cys) in the treatment of Swiss mice with epidermoid skin cancer. Methods. Post test only control group design in 12 Swiss mice, who were successfully initiated with 7,12-dimethylbenzanthracene (DMBA) and promoted with 12-O-tetradecanoyl phorbol-13-acetate (TPA) were randomly allocated into 4 groups: without treatment, treated with PM, treated with Pacli-Cys ,and treated with Pacli-Cys + PM. Tumor removed and prepared for pathologic examination using Mouse Monoclonal Ig G IFN-γ for imunohistochemistry staining, and hematoxylin eosin staining. Result. Supplementation of PM and or Pacli-cys didn’t increase expression of IFN-γ compared with without treatment group . Supplementation of PM increase expression tumor-infiltrating mononuclear cell compared with without treatment group (p=0,05). Suplementation Pacli-Cys significantly increase expression of tumor- infiltrating mononuclear cell compared with without treatment(p=0,05). Supplementation Pacli-Cys + PM significantly increase tumor-infiltrating mononuclear cell compared with without treatment group (p=0,05). The correlation between the expression of IFN-γ and tumor-infiltrating mononuclear cell was significantly positive (p = 0,033 dan r = 0,616). Conclusion. PM and or Pacli-Cys supplementation has no effect on stimulating Interferon-γ production. PM and or Pacli-Cys supplementation has synergic effect on stimulating tumor-infiltrating mononuclear cell. PM supplementation increasing tumor-infiltrating mononuclear cells higher than Pacli-cys. PM+Pacli-Cys did not increase tumor-infiltrating mononuclear cell compare with PM alone or Pacli-Cys alone.
