Abstract

Background: Haemophilus influenzae is one of the important causes of pneumonia and meningitis which is fastidious or different to cultivate. Chocolate agar from sheep blood is the gold standard for cultivating this bacteria. Logistic and financial limitation for the availability of sheep blood in the tropical countries inhibits many laboratories to cultivate H. influenzae. While, human blood which is cheap and easy to provide is not recommended for this research. This research aimed to find an alternative media made from human blood with modification in agar base using tryptic soy agar (TSA). Methods: This research has a true experimental post test only study. Eleventh strains were cultivated on sheep blood derived chocolate agar (SCA), human blood derived chocolate agar (HCA), and TSA modified human blood derived chocolate agar (HCATSA). Growth was evaluated on 24 and 48 hour based on diameter of the colonies, colony count, and characteristic of the colonies. In addition the strains were spiked into sputum and cultured on the same types of media. Growth was evaluated based on diameter and characteristic of colonies. Result: After 24 hour, diameter of colonies from pure suspension on SCA and HCATSA did not differ significantly (p>0,05), while that on HCA was significantly smaller (p=0,000). On 48 hours, diameter of colonies on HCATSA was significantly bigger than that on SCA and HCA (p<0,05). The colonies of H. influenzae which were spiked to sputum on HCATSA were significantly bigger than that on HCA and SCAboth on 24 and 48 hours evaluation (p<0,05).
