EFEK FLUVASTATIN TERHADAP SELISIH JUMLAH LEUKOSIT, NEUTROFIL, DAN ALKALI FOSFATASE SERUM PADA TIKUS WISTAR SEBELUM DAN SESUDAH PAPARAN ASAP ROKOK

Abstract

Background: Exposure to cigarette smoke increases leukocyte counts and ALP levels. Statins reduce leukocytes counts in rat sepsis model. However, statins also have pro-inflammatory effects by increasing TGF-β. This study aims to determine the effects of fluvastatin on delta changes of total counts of leukocyte, neutrophil, and ALP levels before and after exposure to cigarette smoke. Method: This study was a pre and post test only control group design. Rats were divided randomly into 3 groups each 6 rats. Group K: without fluvastatin; P1: fluvastatin administered 5 mg/KgBW; P2: fluvastatin 10 mg/KgBW. Measurement of leukocyte, neutrophil counts, and levels of ALP were performed before and after exposure to cigarette smoke. The number of leukocytes and neutrophils in the blood were measured with blood analyzer. ALP levels in serum were checked by means of Cobas mira. Data were analyzed by paired T test, one way Anova and Kruskal Wallis test. Results: Delta number of leukocytes in group K, P1, and P2 were: 7820; 5817; 7650 respectively (p> 0.05). Delta number of neutrophils in group K, P1, and P2 were: 6340; 4183; 5350 respectively (p> 0.05). Delta ALP levels in group K, P1, and P2 were: 226.80; 180.17; 378.0 respectively with p> 0.05 between groups K vs. P1 and p <0.05 for groups P2 vs K dan P2 vs P1. Conclusion: Delta in counts of leukocyte, neutrophil, and ALP in group P1 are not significantly lower than group K. ALP levels were higher in group P2.
