Abstract

L-Asparaginase merupakan enzim hidrolase yang bekerja mengkatalisis reaksi hidrolisis L-asparagin menjadi amonia dan asam aspartat. L-Asparaginase memiliki peran penting dalam bidang medis, khususnya untuk pengobatan penyakit kanker. Aspergillus niger merupakan salah satu mikroorganisme yang memiliki potensi sebagai somber L-asparaginase. Biotin adalah vitamin yang larut &lath" air dan berfiuigsi untuk tneriabatitu iiieriingkatkari sel. Penelitian ini bertujuan untuk mengetahui pengaruh penambahan biotin pada media pertumbuhan Aspergillus niger terhadap produksi sel Aspergillus niger dan aktivitas spesifik L-asparaginase. Penelitian ini diawali dengan penanaman jamur Aspergillus niger dalam media agar miring (Taoge Ekstrak Agar/TEA), kemudian Aspergillus niger dipindahkan pada media pertumbuhan (Czapek cair) dengan penambahan biotin yang konsentrasinya 0,1 mg/L. Selanjutnya berat biomassa dan aktivitas L-asparaginase ditentukan untuk mengetahui pengaruh penambahan biotin terhadap berat biomassa Aspergillus niger dan aktivitas spesifik L-asparaginase, sedangkan untuk mengetahui pengaruh penambahan biotin terhadap produksi L-asparaginase dapat diketahui dengan cars menumbuhkan Aspergillus niger pada media Czapek cair (tanpa dan dengan penambahan biotin) dengan jumlah media yang Iebih besar. menunjiikkaii Waktii iiikiiba.§i untuk menghasilkan berat biomassa Aspergillus niger dan aktivitas spesifik :kasparaginase adalah_ pada jam ke-48dan fraksi 2 (F2 dengan tingkat kejenu4n 20-40 %) merupakan fraksi yang memiliki aktivitas spesifik tertinggi yaitu 49,391 unit/mg protein. Penambahan biotin 0,1 mg/L pada media pertumbuhan Aspergillus niger dapat meningkatkan berat biomassa Aspergillus niger hingga 40,17 % dan aktivitas spesifik L-asparaginase hingga 73,24 %. LAsparaginase is a hydrolase enzyme that catalyzes hydrolysis reaction of L-asparagin producing ammonia and aspartic acid. Asparaginase has significant role in medical, especially for cancer treatment. Aspergillus niger is one of microorganisms which has potential source for L-asparaginase. Biotins are vitamins soluble in water and have function to facilitate the increasing cell groWth. This research has purpdses to know the influences Of biotin addition into growing medium of Aspergillus niger to its cell production and L-asparaginase's specific activity. This research was begun by the planting of Aspergillus niger on agar slope medium (bean sprouts extract agar), then Aspergillus niger was removed to growing medium (Czapek liquid medium) accompanied by addition of biotin with concentration of 0;1 mg/L. After that the biomass weight and activity of L-asparaginase were deterthined in Order to know the influences Of biotin addition to biomass weight of Aspergillus niger and specific activity of L-asparaginase, while to know the influences of biotin addition to the production of asparaginase might be observed by mean growing Aspergillus niger on liquid Czapek medium (with and without addition of biotin) in larger amount of medium. The results of the research showed that the optimum incubation time for producing biomass weight of Aspergillus niger and specific activity of L-asparaginase was at the 48th hour and fraction 2 (F2 with saturation 20-40 %) was the fraction which had the highest specific activity of 49;391 unit/mg protein. Addition of biotin 0,1 ing/L into growing medium of Aspergillus niger might increase biomass weight of Aspergillus niger until 40,17 % and specific activity of L-asparaginase until 73,24 %.
