Abstract

Bengle (Zingiber cassumunar Roxb.) termasuk salah sate tanaman obat yang turnbuh di Asia Tenggara, khususnya Indonesia. Bengle termasuk famili Zingibera¬ceae yang pada umumnya mengandung senyawa golongan terpenoid dan fenolik. Pada penelitian sebelumnya telah dilakukan identifikasi kandungan kimia rimpang bengle dari ekstrak metanol dan aseton, sedangkan penelitian terhadap ekstrak etil asetat belum dilakukan. Senyawa golongan flavonoid dikenal sebagai anti oksidan alami. Rimpang bengle diketahui mengandung senyawa flavonoid. Oleh karena itu, pada penelitian ini dilakukan identifikasi flavonoid yang terkandung dalam ekstrak etil asetat rimpang bengle dengan spektroskopi UV-Vis dan menguji aktivitasnya dengan metode "Brine Shrimp Letality Test". Ekstraksi dilakukan dengan cara maserasi menggunakan pelarut etil asetat menghasilkan ekstrak kasar. Uji golongan dilakukan untuk mengetahui kandungan kimia. Pemisahan komponen dilakukan dengan metode kromatografi kolom vakum menggunakan campuran pelarut n-heksan dan etil asetat (1 : 1) dan TLC preparatif menggunakan pelarut metanol. Kemurnian senyawa dianalisis dengan TLC dua dirnensi menggunakan pelarut kloroform dan etil asetat. Analisis struktur flavonoid dilakukan dengan spektroskopi UV-Vis menggunakan pelarut metanol p.a dan pereaksi geser untuk menentukan pola oksigenasinya. Pereaksi geser yang digunakan adalah larutan NaOH 2 N, AlC13, dan HCI, serta serbuk NaOAc, dan H3B03. Pita serapan UV-Vis senyawa dalam pelarut metanol p.a berada panjang gelombang 267,5 nm (pita II) dan 335 nm (bahu) (pita I). Senyawa flavonoid hasil isolasi termasuk jenis isoflavon. Senyawa tersebut diperkirakan adalah irigenin Berdasarkan metode "Brine Shrimp Letality Test" senyawa tersebut mempunyai harga LC50 sebesar 91,10 ppm dan berpotensi sebagai obat (anti bakteri). Bengle (Zingiber cassumunar Roxb.) is one of herbal plants growing in South East Asian, especially Indonesia. It is a member of Zingiberaceae family which commonly contains terpenoid and phenolic compounds. Previous research has identified chemical compounds from methanol and acetone extract of bengle rhizome, but from ethyl acetate extract has not identified yet. Flavonoid was known as a natural antioxidant. Bengle rhizome contains flavonoid. In this research, flavo¬noid compound from ethyl acetate extract of bengle rhizome was identified by UV¬Vis spectroscopy method and examined its activity with "Brine Shrimp Letality Test" method. Extraction was conducted by maseration method using ethyl acetat, obtained dark extract (crude). Phytochemistry screening was done to identified chemical compounds. The dark extract was separated by vaccum column chromatography techniques using n-hexane and ethyl acetat (1 : 1) and preparative TLC using metha¬nol. Purity of to the isolated compound was tested by two dimension TLC. Flavonoid sructure was identified by UV-Vis spectroscopy using methanol as as solvent and shift reagent to determine oxigenation pattern. Shift reagents used were NaOH 2 N, AlC13, and HC1, and NaOAc dan H3B03 powder. The UV-Vis spectra of isolated compound in methanol solvent has major absorbtion peak (Band II) in 267.5 nm and 335 nm (Band I, shoulder). It was con-cluded as isoflavonoid group. It was supposed as irigenin. The activity of isolated compound showed LC50 91.10 ppm in which according to Brine Shrimp Lethality Test it has a possibility as drug (antibacteria).
