Abstract
Aktivitas selulase dapat dihambat oleh beberapa inhibitor diantaranya Ag(I), 1-1DTA, Cu(II), dii. Prosentase penghambatan EDTA dan Cu(II) terhadap aktivitas selulase rayap (Reticulitertnes flavipes) pada hidrolisis CMC dapat diketahui dari perfurunan unit aktivitas spesifik selulase. Unit aktivitas selulase (mmol.menit-1.mL-1) ditentukan berdasarkan glukosa yang terbentuk dari reaksi hidrolisis CMC oleh selulase rayap. Produk glukosa ditentukan dengan inetode Nelson-Sdinougyi dan kadar protein ditentukan dengan metode Lowry. Hasil penelitian menunjukkan bahwa Cu(II) dengan konsentrasi 5,0 mM dapat rnenghambat kerja enzim selulase rayap (Reticulitermes flavipes) sebesar 73,25 % dan EDTA dengan konsentrasi yang sarna dapat rnenghambat sebesar 82,44 %. Cellulase activity can be inhibited by several inhibitors, for example Ag(I), EDTA, Cu(II), etc. Percent of inhibition of Cu(II) and EDTA at termites (Reticulitermes flavipes) cellulase activity in CMC hidrolysis showed as spesific activities of cellulase. Cellulase activity (mmol.minutes-l.mL-1) was determined from glucose product in CMC hidrolysis by termites (Reticulitermes flavipes) cellulase. The glucose product was analized with Nelson-Somougyi methode and the enzyme concentration was determined by Lowry methode. The research showed that Cu2+ with concentration of 5.0 mM can inhibited 73.25 % spesific activity of Reticulitennes Avipes cellulase and EDTA with same concentration can inhibited 82.44 %. 1 t ix This document is Undip Institutional Repository Collection.
