Abstract
Telah dilaporkan bahwa tanaman Sidaguri (Sida rhombifolia Linn.) mengandung senyawa aktif dan dapat digunakan sebagai bahan obat. Pada penelitian ini dilakukan identifikasi dan uji aktifitas senyawa dari akar Sidaguri. Serbuk akar Sidaguri dimaserasi dengan pelarut n-heksan sehingga didapatkan ekstrak kasar. Pemisahan senyawa dilakukan dengan kromatografi kolom yak= dengan eluen campuran n-heksan, kloroform, etii asetat (4 : 2 : 1). Fraksi teraktif kemudian diekstraksi dengan campuran pelarut metanol dan n-heksan (1 : 1) dan dipisahkan dengan kromatografi lapis tipis preparatif menggunakan eluen campuran kloroform dan etil asetat (3 : 1). Uji aktifitas dilakukan dengan mengacu pada metode Brine Shrimp Lethality, sedangkan analisis struktur senyawa dilakukan dengan metode GC-MS. Menurut data GC-MS, diperkirakan senyawa yang diperoleh dari KLT preparatif adalah heptadeka-7,13-dien-7-ol; 7-metil-oktadeka-6,14-dien-6-ol; dan 4-kloro-5-etoksi-l-tridek-9-en-4-urfiloksi-4-penten-1-ol. Uji aktifitas terhadap basil KLT preparatif menunjukkan harga LC50 sebesar 17,6058 ppm, yang dapat digolongkan sebagai zat yang bersifat sitotoksik It has been reported that Sida rhombifolia Linn, has active compounds and can be used as the substance medicinize. This research has been done to identificated active compounds and tested the activity from roots of Sida rhombifolia Linn. The powder of roots of Sida rombifolia Linn. were maserated using n-hexane as solvent, obtained crude. Compounds separation were conducted by vaccum column chromatography using mixture of n-hexane, chloroform, ethyl acetic (4 : 2 : 1) as eluent. The most active fraction was extracted with mixture of methanol and n-hexane (1 : 1) and separated by TLC preparative using mixture of chloroform and ethyl acetic (3 : 1) as eluent. Activity test was done by related on method of Brine Shrimp Lethality, while analysis of compounds structure were conducted by GC-MS. According to data from GC-MS, estimated that compounds obtained from TLC preparative are heptadeca —7,13-dien-7-ol; 7-methyl-6,14-octadecadien-6-ol; and 4-chloro-5-ethoxy- 1 -tridec-9-en-4-ynyloxy-pent-4-en-1-01. The activity test to the result of TLC preparative showed. LC50 17.6058 ppm, which could be classified as cytotoxic agent.
