Abstract
Telah dilakukan penelitian terhadap aktivitas spesifik heksokinase dari limbah fermentasi alkohol. Spektrofotometri tampak digunakan untuk menentukan juinlah unit aktivitas heksokinase dan kadar protein Lowry. Aktivitas spesifik heksokinase diperoleh dengan membandingkan unit aktivitas enzim dengan kadar protein larutan sampel. Berdasarkan analisis dari data yang diperoleh, besarnya aktivitas spesifik heksokinase dari tahap terakhir dalam preparasi enzim (larutan untuk kristalisasi) adalah 137,15 unit/mg [0-40 % (NH4)9SO4] dan 66,85 unit/mg [40-55 % (NH4)2SO4] untuk ulangan pertama. Untuk ulangan kedua 146,37 unit/mg [0-40 % (NH4)2SO4] dan 58,77 unit/mg [40-55 % (NH4)2SO4]. Dengan mengetahui aktivitas spesifiknya menunjukkan bahwa dalam limbah fermentasi alkohol terdapat enzim heksokinase, sehingga limbah fermentasi yang tidak, digunakan lagi bisa dimanfaatkan sebagai sumber enzim (heksokinase). It had been researched the specific activity of hexokinase from alcohol fermentation waste. The visible spectrophotometry was used to measure activity units of hexokinase and Lowry protein. The activity specific of hexokinase had been obtained with comparation activity units of enzyme with sample of protein solution. The result of analysis from data were obtained, specific activity of hexokinase from final step in preparation of enzyme (solution for crystallization) was 137,15 unis/mg [0-40 % (NH4)2SO43 and 66,85 units/mg [40-55 % (NH4)2SO4] for one experiment. For second experiment 146,37 units/mg [0-40 % (NH4)9SO4] and 58,77 units/mg [40¬55 % (NH4)2604]. The specific activity showed that in alcohol fermentation waste found hexokinase enzymes, therefore fermentation waste that was not needed could be used as source of enzymes (hexokinase)
