Abstract

Backgrounds: Proinflammatory cytokine is a mediator of inflammation affecting the increase of iNOS and NO release. Vitamin C, a potent antioxidant, is thought to supress the production of inflammatory cytokine, which may result in the inhibition of NO production. Objectives: to know the effect of administration of vitamin C with the dose of 0.52 mg, 1.04 mg and 2.6 mg daily on NO in mice injected with intraperitoneal lipopolysaccharide endotoxin. Methods: the study was a laboratory experimental study with a randomized post test only controlled group design of 20 Balb/c mice injected with intraperitoneal lipopolysaccharide. The mice were then randomly divided into 4 groups, i.e. P1 as a control group, P2 given with 0.52 mg vitamin C daily, P3 given with 1.04 mg vitamin C daily, and P4 given with 2.6 mg vitamin C daily. The NO was obtained from an intraperitoneal macrophage culture 6 hours after the vitamin C administration. Results were then assessed with a nonparametric-statistical test, Kruskal-Wallis test followed with a hypothetical test Mann-Whitney with the degree of significance of p<0,05. Results: The mean NO concentration in P1(18,49±0,727), P2(15,65±0,021), P3(15,52±0,02) and P4(15,47±0,009). There was a significant difference in P2, P3 and P4 compared to P1, p=0,009. There were also significant differences between P2 and P3, p=0,009; P2 and P4, p=0,009; and P3 group and P4 group, p=0,009. Conclusions: Vitamin C could significantly lower the NO concentration of intraperitoneal macrophage in mice exposed to LPS. Intravenous vitamin C with the dosage of 2,6 mg/day decreased the NO intraperitoneal macrophage the most. Keywords: lipopolysaccharide, vitamin C, nitric oxide (NO).
