Abstract

ABSTRACT Background: Aluminum in Tawas cause cell damage kidney epithel tubulus and interference process hematopoiesis. Heavy metals, including aluminum bound protein metalotionin obtained through suplementation zinc. Objective of this study influence of suplementation zinc 0.2 mg, 0.4 mg, 0.8 mg of the major damage to kidney cells epithel tubulus and due process of hematopoiesis in tawas white rat Rattus nurvegicus. Method: Design research is experimental Randimized Post-test Control Group Only. Twenty-four rats Rattus nurvegigus age of 15 days normal body weight, from the Animal Experiment Unit Development, Gadjah Mada University, Yogyakarta divided the 4 groups: group I, II, III and IV. All groups were given Tawas 4%, group II, III and IV plus zinc supplements respectively 0.2 mg / day / head, 0.4 mg / day / head and 0.8 mg / day / head for 30 days. End of the treatment of blood serum was taken for the interference process hematopoiesis and kidney organ to find out the number of kidney cells epithel tubulus the degeneration and necrosis. The statistical analysis using Anova test to find the differences between the four groups and followed by the Bonferroni test for knowing the significance level in each group is different when compared with the control group. Results: There is no difference result on supplementation zinc 0.2 mg, 0.4 mg and 0.8 mg of cell epithel tubulus kidney degeneration and there is a meaningful difference to the cell necrosis. There are meaningful differences in supplementation zinc 0.2 mg, 0.4 mg and 0.8 mg of Hb, Ht, the number of erytrocyte and retyculocyte. Conclusion: Suplementation zinc 0.2 mg, 0.4 mg and 0.8 mg necrosis affect the number of cells epithel tubulus kidney and the process of hematopoiesis. Keywords: Tawas, Zinc, Hematopoiesis, kidney tubulus ABSTRAK Latar Belakang: Alumunium dalam tawas menimbulkan kerusakan sel epitel tubulus ginjal dan gangguan proses hematopoiesis. Alumunium termasuk logam berat diikat protein metalotionin diperoleh melalui suplementasi seng. Penelitian bertujuan membuktikan pengaruh suplementasi seng 0,2 mg, 0,4 mg, 0,8 mg terhadap hambatan kerusakan sel epitel tubulus ginjal dan proses hematopoiesis akibat pemberian tawas dalam pakan tikus putih Rattus nurvegicus . Metode: Desain penelitian eksperimental laboratorium menggunakan Randomized Post test Control-Group Only. Duapuluh empat tikus Rattus nurvegigus umur 15 minggu berat badan normal, dari Unit Pengembangan Hewan Percobaan, Universitas Gadjah Mada, Yogyakarta dibagi 4 kelompok yaitu: kelompok I,II,III dan IV. Semua kelompok diberi tawas 4%, kelompok II,III dan IV ditambah suplemen seng berturut-turut 0,2 mg/hari/ekor, 0,4 mg/hari/ekor dan 0,8 mg/hari/ekor selama 30 hari. Akhir perlakuan diambil serum darah untuk mengetahui gangguan proses hematopoiesis dan organ ginjal untuk mengetahui jumlah sel epitel tubulus ginjal yang mengalami degenerasi dan nekrosis. Analisis uji statistik menggunakan uji Anova untuk mengetahui perbedaan keempat kelompok dan dilanjutkan uji Bonferroni untuk mengetahui tingkat kemaknaan pada masing-masing kelompok yang berbeda bila dibandingkan dengan kelompok kontrol. Hasil penelitian: Tidak terdapat perbedaan suplemen seng 0,2 mg, 0,4 mg dan 0,8 mg terhadap degenerasi sel epitel tubulus ginjal dan terdapat perbedaan bermakna terhadap sel nekrosis. Terdapat perbedaan bermakna suplemen seng 0,2 mg, 0,4 mg dan 0,8 mg terhadap Hb, Ht, jumlah eritrosit dan retikulosit. Simpulan: pemberian suplementasi seng 0,2 mg, 0,4 mg dan 0,8 mg berpengaruh terhadap jumlah nekrosis sel epitel tubulus ginjal dan proses hematopoiesis.
