Abstract

Background: Andrographis paniculata (A. paniculata) can stimulate immune response of BALB/c mice during the infection of Plasmodium berghei ANKA (P. berghei ANKA). While interferon-gamma (IFN- γ) has been proved to be protective during the infection. Yet there is no proof that the protective mechanism of the herb is mediated by IFN- γ. The objective of this study was to find out whether A. paniculata has an effect on IFN- γ producing capacity in mice inoculated by P. berghei ANKA. Methods: This study was pure experimental with post test only control group design. It used 36 BALB/c mice, divided randomly into treated and control groups. Each mice in treated group received 16,608 mg methanol extract of A. paniculata, solved in CMC-Na 0,5%, started from 7 days pre-inoculation until 8 days post-inoculation (p.i.). While control group received CMC-Na 0,5% 0,1 ml/day/mice with same duration. All of the mice was inoculated with P. berghei ANKA on day 7 after the treatment started. IFN- γ producing capacity was measured on day 3, 6, 8 p.i. Data was processed using Kruskal Wallis and Mann Whitney statistical analysis test. Results: IFN- γ producing capacity of treated group was significantly higher than control group on day 3 p.i. of P. berghei ANKA. In treated group, there was significant difference of IFN- γ concentration between day 3 and 6 p.i. In control group, there were significant differences of IFN- γ concentration between day 3 and 6, and also day 3 and 8 p.i. of P. berghei ANKA. Conclusion: A. paniculata herb extract increase spleenocytes’ IFN- γ producing capacity on day 3 p.i. of P. berghei ANKA. Keywords: Andrographis paniculata, IFN-γ, Plasmodium berghei ANKA
