Abstract

Background: Pineapple (Ananas comosus (L.) Merr) is a traditional herbal medicine which has some effects of anti inflamation, anti oxidant, anti cancer, anti bacterial and anti fungal. Zinc pyrithione is a substance that is used as anti bacterial, anti fungal, and anti seborrhea. Ketoconazole works for blocking ergosterol synthesis, an important component for membrane integrity of fungal cell. Dandruff is excessive scales production of the scalp without signs of inflamation. Pityrosporum ovale is human’s skin normal flora, but in people with dandruff, the number of P. ovale increases. Objective: To prove the effectiveness of 100% squeezed pineapple, 1% zinc pyrithione and 1% ketoconazole in vitro to the growth of P. ovale in dandruff. Method: This research was using an experimental study. As samples were 30 patients of dandruff with clinical founding. Diagnosis of P. ovale was based on the result of microscopic examination of skin scrapping from samples using KOH 10% with blue black parker ink, and inoculation on the olive oil Sabouraud Dextrose Agar with chloramfenicol 50μg/cc media on 370 C for 5 days. The colonies of P. ovale were diluted in sterilized 0,9% NaCl to make the solution equal to 0,5 McFarland standard. As many as 0,1 cc of solution was cultivated on the olive oil SDA media supplemented with 100% squeezed pineapple, 1% zinc pyrithone and 1% ketoconazole. After that, the media were incubated on 370 C for 5 days. The difference proportion of growth was analyzed by chi square test with degree of significance of p<0,05. Result: 30 media of olive oil SDA contained 100% squeezed pineapple, 30 (33,3%) were found (+)/positive for P. ovale growth and none(0%) were found (-) /absence for P. ovale growth. 30 media contained 1% zinc pyrithione, 6(6,7%) were found positive and 24(26,7%) negative. 30 media contained 1% ketoconazole, 5(5,6%) were found positive and 25(27,8%) negative The result of chi square test is significant (p=0,000). Conclusion: The effectiveness between 100% squeezed pineapple, 1% zinc pyritihione and 1% ketoconazole in inhibiting the growth of P. ovale in dandruff is significantly different.
