Abstract

Background: Gorgonian Isis hippuris contains hippuristanol, an anti cancer compound, but there has been no in vivo toxicity study. This study was aimed to know the effects of subchronic administration of gorgonian Isis hippuris extract on the liver histopathology of Swiss mice. Methods: This research was designed as a randomized post test only control group, using 25 male and 25 female Swiss mice that fulfilled the inclusion criteria. Experimental mice were treated by 3 levels ethanol extract of gorgonian Isis hippuris i.e. 0,15 mg (P1), 1,5 mg (P2) and 15 mg extract (P3) per 0,5 g standard pelleted diet daily. Two control groups consisted of mice fed the standard diet only (Cneg) and mice fed pelleted diet that contain ethanol (C0). After 3 months of extract administration, all mice were terminated and their liver were processed and stained by hematoxylin eosin. Liver histopathology was examined using modified Knodell score. Results: Liver samples of all groups showed low level of damage, histopathology index between 0,04-0,14. The highest median i.e. 0,095 (0,088; 0,103) in P2 and the lowest i.e. 0,060 (0,055; 0,085) in C0. Kruskal-Wallis test showed no significant difference between liver histopathology index of experimental and control groups (p=0,085). Conclusion: Gorgonian Isis hippuris extract orally administered has no effect on the liver histopathology which indicates its safety for short to medium term oral use in mice.
