Abstract

Background : Diabetes mellitus can depress reproduction hormones, including testosterone. This condition may influence motility maturation of the sperm in the testes. The previous studies had reported that Nigella sativa had effect in increasing testosterone. A study was carried out to know the effect of Nigella sativa on sperm motility in alloxan-induced diabetic mice. Method : This was a laboratory experimental study used Post Test-Only Control Group Design. Samples were 30 Balb/c male mice, which were divided into five groups randomly, each group contained 6 mice. Group K- (negative control) was untreated group. Group K+ (positive control), P1, P2, and P3 (treated group) were given a single intraperitoneal injection of alloxan with dosage 3,75 mgs on 1st day. On 4th day until 15th day, group P1, P2 and P3 were given Nigella sativa oil everyday. The dosage for each group was 0,1 ml, 0,2 ml and 0,3 ml. On 16th day all mice were sacrificed and the sperm motility analysis was performed. Result : Based on examination in sperm, it was found that motility of sperm in group K+ was also significantly lower than K- (p=0,011). Motility of sperm in group K+ was significantly lower than P3 (p=0,027). Motility of sperm in group K- was significantly lower than P1 (0,019). Data analyzed between group P1 and P3 showed significant difference (p=0,044). And data analyzed between group P1 and P2 showed no significant difference (p=0,406) as well as between group P2 and P3 (p=0,715). Conclusion : Nigella sativa oil supplementation in 0,3 cc dosage can increase motility of sperm in alloxan-induced diabetic mice. Keywords : diabetes mellitus, testosterone, Nigella sativa
