Abstract
Background: Mononuclear cell macrophage plays an important role as phagocyte and antigen-presenting cell (APC) for a specific immune system initiation that can be improved by the treatment of immunomodulator. Tolak Angin Anak Cair (TAAC), usually targeted for children, is a standardized herbal drug developed from Tolak Angin Cair (TAC) with an addition of buffalo-horn extract as antipyretic. TAC has been proven to improve peripheral T cell and Interferon-gama (IFN-γ) as the activator of macrophage. This study was designed to observe how the macrophage’s phagocytosis index on Swiss mice can be affected by an increasing dosage of TAAC. Method: Pure experimental study laboratory with Post Test Only Control Group Design was performed. Forty Swiss mice weighing 25-35 grams of 12-week old were equally divided into four groups: K (no TAAC administration), P1 (TAAC 0.125ml/day), P2 (TAAC 0.25ml/day), and P3 (TAAC 0.5ml/day). Interventions were given for seven days. Macrophages were isolated directly from peritoneal’s macrophages of mice. The data were then analyzed by Kruskal-Wallis and Mann Whitney tests. Result: Groups treated with TAAC showed a significant increased of macrophage’s phagocytosis index compared to control group (p=0.004). The treatment of TAAC that gave significant result was P2 group (0.25ml/day). Comparison between the treatment groups did not show any significant difference, except between P2 and P3 groups. Conclusion: Administration of TAAC has been proven to show a significant increased effect on macrophage’s phagocytosis index. However, the macrophage’s phagocytosis index was not directly proportional to the increasing treatment of TAAC; the optimum dosage was reached at 0.25ml/day.
