Abstract

Background: Dextromethorphan is an opioid antitussive that can be obtained and used freely thus increased the risk of abuse and poisoning. The use of high doses of dextromethorphan can cause destruction in the central nervous system and the destruction to other organs, liver is one of them. This experiment’s objective is to observe the effect of per oral gradual doses dextromethorphan to the histopatology image of Wistar rat’s liver. Methods: This laboratory experimental study with post test only control group design used 24 Wistars which divided randomly into 4 groups. Each group consists of 6 Wistars. Control group (K) given no intervention. P1 group given dextromethorphan per oral 0,175 mg/gBW, P2 group given 0,35 mg/gBW, and P3 group given 0,7mg/gBW. Wistar rat’s that died were taken the liver while not dead until 7 days were decapitated and then samples of liver tissue were made. Data measured were the grade of hepatocytes destruction to assess the histopatology image of Wistar rat’s liver. Result: The highest liver histopathological score was in P2. The score evaluated parenchymatous degeneration, hydropic degeneration, and necrosis. The Anova test showed significant difference (p=0,000). Continued with Post Hoc test that showed significant difference in K-P1(p=0,027), K-P2(p=0,000), K-P3(p=0,000), P1-P2(p=0,000), and P1-P3(p=0,003). There was no significant difference in P2-P3(p=0,299). Conclusion: There was effects of dextromethorphan 0,175 mg/gBW, 0,35 mg/gBW, and 0,7 mg/gBW to the histopatology image of Wistar rat’s liver in which happened parenchymatous degeneration, hydropic degeneration, and necrosis. There was a significant difference of histopatology image of Wistar rat’s liver between control group with treatment group, P1 with P2, P1 with P3. There was no significant difference of histopatology image of Wistar rat’s liver between P2 with P3. Keywords: dextromethorphan, liver’s histopatology image
