Abstract
Penelitian untuk membuktikan keuntungan dan resiko konsumsi harian fitoestrogen pada proses reproduksi mencit telah dilakukan pada bulan April sampai dengan November 2005. Penelitian menggunakan mencit betina dewasa dengan berat 25 — 30 gram sebanyak 32 ekor. Hewan uji dikelompokkan menjadi kelompok kontrol (diberi akuades) dan kelompok perlakuan (diberi bahan uji). Bahan uji yang diberikan berupa tepung kedelai yang diberikan secara oral menggunakan jarum gavage dengan pelarut akuades. Tepung kedelai dengan dosis 0,975 mg/ekor/hari diberikan selama 48 hari. Penentuan siklus estrus dilakukan dengan pembuatan apus vagina dengan pewarnaan Papanicolau. Sampel darah untuk pengukuran hormon diambil pada setiap tahapan siklus estrus, sesuai metoda Oduma et al. (1995). Penentuan kadar hormon estradiol dilakukan dengan ELISA Kit. Data dianalisa dengan Student T-test. Basil pengukuran kadar hormon estradiol menunjukkan kadar yang lebih rendah pada kelompok perlakuan tetapi tidak ada perbedaan yang nyata dengan kelompok kontrol. Pola fluktuasi hormon menunjukkan gambaran hiperbola yang spesifik pada mammalia, kadar tertinggi pada fase proestrus, menurun pada fase estrus, dan kadar terrendah pada fase metesrus dan diestrus. Berdasar hasil penelitian tersebut dapat disimpulkan bahwa konsumsi harian tepung kedelai tidak menyebabkan perubahan pola fluktuasi tetapi berpotensi menekan sintesa atau aksi hormon estradiol path proses reproduksi. Effect of fitoesterogen daily consumption on mice reproduction has been investigated since April to November 2005. This research uses 35 mice, with the weight ranges between 25 to 30 gram. The mice were grouped into controls (treated by aquades), while another group was treated by soybean flour. As much as 0.975 mg of soybean powder in aquadest solution was given orally to each mouse per day, using gavage spuit. This experiment was conducted during 48 days. The estrus cycle was decided by vagina smear, followed by Papanicolau staining. Blood sample for hormone measurement was taken every estrus cycle phase, as done by Oduma et al. (1995) method. Measurement of estradiol concentration was done by ELISA kit. The data were analysed by Student t-test. This experiment resulted that treated mice have lower estradiol concentration than the control group. However, the different was not significant. The hormone fluctuation pattern indicated specific hyperbole on mammals. The highest concentration resulted in proestrus phase, decreasing in estrus phase, and the lowest concentration is in metestrus and diestrus. Based on these results, it can be concluded that daily consumption of soybean flour could not affect fluctuation pattern, but it potentially inhibit synthesis or estradiol action in reproduction process.
