Abstract

Background: Synthetic glucocorticoid dexamethasone is used widely in the community, but the use of large doses in the long term can affect the liver. The liver is a place where drugs and other toxic substances are metabolized, however the absence of studies specifically examine the effect of dexamethasone on liver histology image. The prove the effect of giving rise per oral dose of dexamethasone 30 days on Wistar rat liver histologic picture. Method: Experimental research design is the post test only control group design. The sample of 20 male wistar rats were divided into four groups randomly, K is the control group, were given only distilled water. P1 group were given dexamethasone orally with sonde 0.018 mg / day, Group P2 given 0.73 mg / day, the P3 group was given 1.44 mg / day. After 30 days mice were then terminated and the liver was taken to make preparations. Preparations then read under a microscope. Target that is read is the change in histological structure of liver cells in five fields of view by using the Roenigk score Spoiled integrity. Result: The highest liver histological score was in P3 group. The score evaluated parenchymatous degeneration, hydropic degeneration, and necrosis. The Anova test showed significant difference (p=0,0001). The Post Hoc test showed significant difference in K-P1(p=0,0001), K-P2(p=0,0001), K-P3(p=0,0001), P1-P3(p=0,0001), and P2-P3(p=0,0001), and at P1-P2(p=0,0001). Conclusion: Differences in the level of liver cell damage in the group given dexamethasone than in control group. Differences in the level of liver cell damage also occurred between the treatment group, where the level of liver cell damage P3 greater than P2 groups and group P2 greater than P1 group. At P3 group found that mean liver damage cell necrosis, hydropic degeneration group P2 form of degeneration parenkimatosa P1 group. This shows there is a relationship between increasing doses of dexamethasone on the level of damage to liver cells of Wistar rats. Keywords :Dexamethasone, liver cell damage
