Abstract

Background: Valerian (Valeriana officinalis) is a herbal medicine that has sedative effects which are now widely used to solve sleep disturbances. Valerian is excreted by the kidney, whereas it contains Cd and Pb which are nephrotoxic. The aim of this study was to analyze whether there is any effect on renal microscopic appearances and creatinine concentration after subchronic treatment of valerian. Method: This research was an experimental study using the pre and post test controlled group design and the post test only controlled group design. The samples were 20 male wistar rats, randomly divided into four groups. One control group was given aquadest, and three treatment groups (P1 , P2, P3) were each given a dose of valerian 9 ; 18 ; 36 mg / rat through a stomach sonde once a day for three months. At the time before and the end of treatment,rat’s blood were taken. In third month, the rats were terminated to observe their histopathologhic appearances. Microscopic data, delta pre and post test creatinine, and post test creatinine concentration between groups were analyzed using One-Way Anova. Microscopic data followed by Post-Hoc Test. Result: There were significant differences in renal microscopic appearances among groups, between: K-P1 with p = 0.001, K-P2, P3, P1-P3 with p = 0.000, and P2-P3 with p = 0.006 .However, there were no significant differences in renal microscopic appearances between P1-P2 with p = 0.054. There were no significant differences in pre and post creatinine concentration with p = 0693, and in post test creatinine concentration among groups with p = 0.370 Conclusion : Subchronic treatment of valerian extract affect renal microscopic appearance in which renal damage is still minimal microscopic form of renal tubular narrowing, but no effect on creatinine concentration.
