Abstract

Background:Defibrinated sheep blood agar (DSBA) is standart media for microbiology laboratory test as some countries have difficulty providing it. Therefore, alternative media, such as human blood agar (HuBA), is needed. This study aimed to analyze whether HuBA can be used as alternative media and to compare the growth of S. pneumoniae on DSBAand HuBA. Methods: This study used True experimental design post test only against six strains of S. pneumoniae. Samples were divided into four groups, group 1 and 2 where the pure strains of S. pneumoniae (assessed colony counts, colony size and diameter of hemolysis as dependent variable 1) and groups 3 and 4 which was spiked with sputum (assessed colony size, diameter of hemolysis and typical colony characteristics as a dependent variable 2) and all of them was isolated on DSBA and HuBA (independent variable). Data was described in graphics and analyzed by Kolmogorov-Smirnov test in SPSS 15 for windows. Results: Analysis of dependent variable 1 are colony counts p (24 hours) = 1.000 and p (48 hours) = 1.000, colony size p (24 hours) = 1.000 and p (48 hours) = 1.000 and the diameter of hemolysis p (24 hours)= 0.441 and p (48 hours) = 1.000. Analysis of dependent variable 2 are colony size p (24 hours) = 1.000 and p (48 hours) = 0.441, diameter of hemolysis p (24 hours) = 0.893 and p (48 hours) = 1.000 and typical colony characteristics p (24 hours) = 1.000 and p (48hours) = 1.000. Conclusion: The growth of S. pneumoniae on DSBA and HuBA both with and without spike with sputum at 24 and 48 hours of observation are not significantly different. Keywords: S.pneumoniae, DSBA, HuBA
