Abstract

Background : Dextromethorphan is an opioid-derived antitussive drug that can be obtained and used freely so the risk of abuse is quite high. Some source mention the usage of dextromethorphan in high dose could cause central nervous system disorders. This study/research aims to investigate the effect of multilevel per oral dose of dextromethorphan on histopathology image of wistar rat brain. Method : This research/study use post test only group design. The number of sample is 24 wistar rats which then divided into 4 groups. Each groups consist of 6 rats. K is control group that is not given any kind of treatment at all. P1 is given destromethorphan per oral ½ x lethal dose, P2 is given per oral lethal dose, and P3 is given per oral 2 x lethal dose. The brain of a dead wistar rat is taken while on the seventh day, alive wistar rat is decapitated, than brain tissue specimen is made. Data that will be observe is the amount of damage of the wistar rat brain cell. Result : The average number of the damaged brain neuron cell in group K = 4, 67, P1 = 38,17, P2 = 134,83, and P3 = 201,00. The estimation include cell necrosis. There is a significant different (p=0,000) found out in One Way Anova test. Which then continued to Post Hoc test whereas there is significant different in K – P1, K – P2, K – P3, P1 – P2, P1 – P3, P2 – P3 (P<0,05). Conclusion : There is an effect by giving dextromethorphan to histopathology image of wistar rat brain. There is a different image of histopathology brain between control group and treatment group, also between each treatment group.
