Abstract

Traditional medicine has been part of Indonesian people daily life for generation. One of such medicine is "benalu teh" (Loranthus sp.). The claim and publication that stated benalu teh is an anti tumor medicinal plants are not substantiated by scientific evidence. It is therefore this study was undertaken to study the anti tumor potency of benalu teh. The present study is designed to demonstrate the anti tumor activity of benalu teh extract. The study tested the hypothesis that benalu teh extract can change the response of tumor cells toward tumor necrosis factor alpha. The hypothesis was tested in vitro by studying the changes of sensitivity of a fibrosarcoma, WEHI-146 cells, toward tumor necrosis factor alpha. Benalu teh extraction was carried out at The Laboratory of Natural Product Chemistry LIPI Bioteknologi Cibinong and the in vitro TNFa toxicity assay was carried out at The Laboratory of Biochemistry Inter-University Centre of Biotechnology Gadjah Mada University. The study showed that S. junghunii dan S. oortiana are capable of modulating the susceptibility of fibrosarcoma to molecules TNFa. Stem extracts of S. junghunii or S. oortiana is more potent in modulating the susceptibily to TNFa compared to leave extracts which is shown by smaller LDa, value of stem extracts. However, comparing stem extracts of S. junghunii to S. oortiana it can be concluded that stem extract S. oortiana is stronger in eliciting increase modulation than S. junghunii. However when comparing leave extracts of the two species, S. junghunii is stronger thant S. oortina. These results further confirmed the possibility of the biological action of benalu teh as an anti tumor was through modulation of susceptibility of tumor cells to TNFa molecules. Obat tradisional telah menjadi bagian tak terpisahkan dari kehidupan keseharian masyarakat. Salah satu contoh obat tradisional adalah "benalu teh" (Loranthus sp.). Klaim/pernyataan yang hidup di masyarakat mengenai benalu teh sebagai obat anti kanker/tumor dan beberapa publikasi yang menyebutkan bahwa benalu teh merupakan obat tumor/kanker belum disertai bukti ilmiah mengenai aktivitas biologisnya maupun kemungkinan mekanisme kerja ekstrak benalu teh. Oleh karena itu penelitian mengenai pengaruh ekstrak benalu teh terhadap sel tumor dilakukan. Penelitian dilakukan untuk menguji hipotesis bahwa ekstrak benalu teh dapat merupah respon sel tumor fibrosarcoma, WEHI-164. Hipotesis ini diuji secara in vitro melalui pengukuran perubahan kepekaan sel-sel tumor fibrosarcoma terhadap molekul tumor necrosis factor alpha (TNFa). Penelitian dilakukan di dua laboratorium yaitu Laboratorium Kimia Bahan Alam LIPI Bioteknologi Cibinong untuk persiapan ekstraksi benalu teh dan laboratorium Biokimia, PAU Bioteknologi UGM untuk pengujian anti tumomya secara in vitro. Hasil penelitian menunjukkan bahwa Daun dan ranting benalu teh S. junghunii dan S. oortiana masing-masing mampu meningkatkan kepekaan sel tumor fibrosarcoma terhadap serangan molekul TNFa. Ekstrak ranting baik pada S. junghunii maupun S. oortiana memiliki LD50 lebih kecil dari LD50 ekstrak daun sehingga ekstrak ranting lebih kuat dalam meningkatkan kepekaan sel tumor fibrosarma terhadap serangan molekul TNFa. Dengan hasil ini maka mekanisme yang mungkin terjadi pada aktifitas anti tumor benalu teh S. junghunii maupun S. oortiana adalah melalui peningkatan kepekaan sel-sel tumor terhadap molekul TNFa yang dilepas oleh sel-sel kekebalan seperti macrophag dan sel-sel-T.
