Abstract
The claim of benalu teh as an anti tumor medicinal plants by the Indonesian people has been substantiated by scientific evidence previously shown by Murwani (2002, 2001a,b). The evidence showed that benalu teh S. oortiana is directly cytotoxic to tumor cells and causing their lysis and capable of sensitizing the tumor cells more susceptible to the molecule TNFa known to be release by effector immune cells. Although the extract is cytotoxic to fibrosarcoma, it does not cause any adverse effect to normal fibroblast cells. This evidence has an important implication for drug development and its use for alleviating cachexia effect of in vivo TNFa therapy on tumor patients. However it is important to elucidate first the active constiuents of the extract responsible for the anti tumor activity and therefore becomes the aim of this study. During the first year of this study two species of benalu teh i.e. S. junghuhnii and S. oortiana could be collected. From the water extract of S. junghunii leaves, 6 fractions eluted through Silica column were obtained. One of this fraction i.e. fraction number 3 were repurified by Silica column and produced 3 fractions designated as fraction 3-1, 3-2, dan 3-3. Fraction 3-2 were further purified by HPLC and the collected pure isolates were subjected to spectral analysis. The results of the spectra identified the chemical compound of fraction 3-2 as caffeine which had the same pattern of spectra as standard caffeine from the Library data. The water extract from S. oortiana stem eluted through Sephadex LH-20 produced 8 fractions. The fraction number 4 was further purified by Silica Column and produced 4 fractions designated as fraction 4-1, 4-2, 4-3, dan 4-4. Fraction 4¬4 was further purified by HPLC to produce 3 chemical compounds. The three compounds were each subjected to UV and IR spectral analysis. From these incomplete spectral analysis it can be concluded that the three coumpounds were each a distinct or different chemical compound. Klaim/pernyataan yang hidup di masyarakat mengenai benalu teh sebagai obat anti kanker/tumor memiliki alasan yang kuat dan hal ini ditopang oleh adanya penelitian sebelumnya oleh penulis yang menunjukkan bahwa benalu teh dapat merusak sel tumor secara langsung serta membuat sel tumor menjadi lebih peka terhadap molekul tumor-necrosis-factor-alpha yang biasanya dilepas oleh sel kekebalan tubuh dalam melawan tumor. Ekstrak benalu teh meski bersifat merusak sel tumor tidak berpengaruh terhadap sel normal. Dan hasil penelitian tersebut yang memiliki implikasi pengembangan obat dari ekstrak air benalu teh maka penelitian untuk mengetahui dan memperoleh senyawa aktif anti tumor dari ekstrak air benalu teh Indonesia yang selama ini belum terungkap dilakukan. Percobaan tahun pertama meliputi dua spesies benalu yaitu S. junghuhnii dan S. oortiana yang terkoleksi pada saat pengambilan sampel dari lapang. Diperoleh 6 fraksi dari isolasi ekstrak air daun S junghuhnii melalui kromatografi kolom silica. Salah satu isolat ini (fraksi 3) dimurnikan kembali dengan kolom silica dan memberikan 3 fraksi yaitu fraksi 3-1, 3-2, dan 3-3. Fraksi 3-2 dimurnikan dengan kromatografi cair kinerja tinggi (HPLC) dan berhasil dimurnikan dan berdasarkan data-data spektra dapat diidentifikasi sebagai kafeina. Isolasi ekstrak air ranting S. oortiana menghasilkan 8 fraksi setelah melalui kromatografi kolom Sephadex LH-20. Fraksi 4 yang dimurnikan lanjut melalui kolom silika menghasilkan 4 fraksi yaitu fraksi 4-1, 4-2, 4-3, dan 4-4. Fraksi 4-4 dimurnikan lebih lanjut menggunakan kromatografi cair kinerja tinggi (HPLC) dart menghasilkan 3 senyawa. Berdasarkan data spektra UV, IR, dan GC-MS ketiga senyawa tersebut dapat diketahui merupakan senyawa yang berbeda satu dengan yang lain.
