Abstract

Background : Acetaminophen is an effective analgesic – antypiretic that is freely distributed and commonly used for self medication. Acetaminophen has since proven to be remarkably safe drug at appropriate dosage, so it is widely sold without receipt. Since the wide availability of the products consequently, overdose, drug abuse and theraupetic misadventure of acetaminophen are common consideration. The most serious adverse effect of acute overdose of acetaminophen is dose dependent. In very high dose, the toxic reactive of acetaminophen metabolites bind to protein components of kidney that induce kidney injury. To determine the severity of kidney damage, can be assessed by glomerulofiltration rate evaluation through measurement of serum ureum and creatinin level. The objective of this experiment was to find out the effect of acetaminophen at variation of dosage to ureum and creatinin serum level of Wistar rat. Method : This experimental study with post test only control group design, used 24 Wistar male rats which were divided into 4 groups. Each group was adaptated for 7 days. On the 8th day group K received acetaminophen 0 mg/KgBW. Group P1 received acetaminophen 1200 mg/KgBW. Group P2 received acetaminophen 2400 mg/KgBW. Group P3 received acetaminophen 4800 mg/KgBW. Retroorbitalis vein blood were taken from all groups on the 12th day and the serum ureum and creatinin level were measured. Results : Outcome of Annova test in serum ureum p=0,000, serum creatinin p=0,000. Outcome of Post-Hoc test in serum ureum were K-P1 (p=0,040), K-P2 (p=0,000), K-P3 (p=0,000), P1-P2 (p=0,045), P1-P3 (p=0,000), P2-P3 (p=0,000), in serum creatinin were K-P1 (p=0,001), K-P2 (p=0,000), K-P3 (p=0,000), P1-P2 (p=0,001), P1-P3 (p=0,000), P2-P3 (p=0,035), The result showed significant differences because p<0,05. Conclusion : There were differences in ureum and creatinin serum level between untreated group and acetaminophen treated group. There were also differences in ureum and creatinin serum level between various dosages of acetaminophen treated group. Ureum and creatinin serum level were rised along with the rising of acetaminophen dosage. Keywords : Serum ureum, serum creatinin, acetaminophen.
