Abstract

Background: Acetaminophen is one of the analgesic and antipyretic drugs derivates from para amino phenol that commonly used in the world. However, nowadays it also commonly used in the cases of suicide attempt. The nefrotoxin metabolites of acetaminophen accumulates in kidney, as a major excretion organ of acetaminophen, especially on proximal tubule, and causes changes on histological appearance. Objective: To know the effect of oral administered in various dosage of acetaminophen on histological appearance from proximal tubules of Wistar Rats kidney. Method : This experimental study used Post Test Only Control Group Design, with 24 male Wistar rats divided into 4 groups, with age of 4,5-6 months and weight of 200-300 gram. K is the control group, without being given any Acetaminophen. P1 is rats being given Acetaminophen orally ½ x 2400mg/ KgW. P2 is rats being given Acetaminophen orally 2400mg/KgW. P3 is rats being given Acetaminophen orally 2 x 2400 mg/KgW. After that the kidney were taken to be observed and made into slides with HE stain, then the proximal tubules were examined. The data were analyzed by the test of differences Kruskal-Wallis and Mann-Whitney with the degree of significance was 5%. Result: The administered of Acetaminophen altering the histological appearance of Wistar rats proximal tubules of kidney which are albuminosa degeneration. Outcome of Kruskal-Wallis test showed significant differences between all groups (p=0,000). The outcome of Mann-Whitney test: between group K and P1 (p=0,004), group K and P2 (p=0,002), group K and P3 (p=0,004), group P1 and P2 (p=0,006), group P1 and P3 (p=0,004), and group P2 and P3 (p=0,032). Conclusion: There were differences of histopathology appearance from proximal tubules of Wistar Rats kidney between untreated group and acetaminophen treated group. There were also differences histopathology appearance from proximal tubules of Wistar Rats kidney between various dosages of acetaminophen treated group, and proximal tubules cell injury was rising along with the rise of acetaminophen dosage.
