Abstract

Background: Methotrexate, an antimetabolit, causes severe side effect such as hepatotoxicity and fibrosis. Alanin aminotransferase (ALT) is a relatively specific indicator of acute liver cell damage. Nigella sativa seeds extract had antioxidant effect. The aim of this study was to prove the ability of Nigella sativa in preventing increase of ALT induced by methotrexate. Method: This study was a randomized controlled trial. The sample were 30 male wistar rats, aged 12-16 weeks, 180-200 grams in weight, divided randomly into 5 groups (K1, K2, P1, P2 and P3). K1 received standard diet only. K2 received 0,54 mg methotrexate orally from 16th-20th day. P1 received 0,54 mg methotrexate orally from 16th- 20th day and 0,008 ml Nigella sativa twice a day from 8th - 21th day. P2 received 0,54 mg methotrexate orally from 16th-20th day and Nigella sativa 0,08 ml twice a day from 8th-21th day. P3 received 0,54 mg methotrexate orally from 16th-20th day and 0,8 ml Nigella sativa twice a day from 8th-21th day . On the 22nd day blood sample was taken from wistar rats’ retroorbital veins to measure the ALT level. Result: The study showed that ALT level among the four groups are not significantly different (p=0,196). Conclusion: Nigella sativa seeds extract at the 0,008 ml, 0,08 ml, and 0,8 ml doses are not proven to prevent mehtotreksat induced ALT level elevation, because the administration of methotrexate did not elevate ALT level.
