PENGARUH PEMBERIAN EKSTRAK BIJI JINTAN HITAM (Nigella sativa) TERHADAP JUMLAH LEUKOSIT TIKUS WISTAR YANG DIBERI METOTREKSAT

Abstract

Background : The side effects of methotrexate such as bone marrow depression could bring into fall in leukocytes count condition. Black seed had been known to has the effect of bone marrow stimulation. The objective of this study is to prove the effect of the oil extract of black seed on leukocytes count of methotrexate-treated rats. Method : This study was an experimental study with Randomized Pre and Post Test Control Group Design. Samples were 28 Wistar rats, divided randomly into 4 groups (control group, first test group, second test group and third test group). All groups were treated with 0,54 mg methotrexate orally on the 16th until 20th day. On the 8th until 21st day, all treatment group were treated with black seed extract twice daily orally at dose 0,008 ml for the first test group, 0,08 ml for the second test group and 0,8 ml for the third test group. Leukocytes were counted on the 7th and 22nd day using Neubauer Improved chamber. The blood samples were obtained from the retro orbital vein. Result : Leukocytes count of the Wistar rats after methotrexate and black seed treatment among the treatment groups was decreased significantly (p < 0,05 on paired t-test). Conclusion : The administration of black seed extract could not increase leukocytes count of methotrexate-treated rats. Keywords : methotrexate, black seed, leukocytes count
