PENGARUH PEMBERIAN EKSTRAK BIJI JINTAN HITAM (Nigella sativa) TERHADAP JUMLAH TROMBOSIT TIKUS WISTAR YANG DIBERI METOTREKSAT

Abstract

Background: Chemotherapy methotrexate often causes bone marrow depression as side effect, which is reflected as thrombocytopenia. Nigella sativa extract was proved that it could increase bone marrow cells proliferation. The objective of this study is to determine the effect of Nigella sativa extract on platelets count of Wistar rats given methotrexate. Methods: This study was an experimental study with Randomized Pre Test and Post Test Control Group Design. Samples consist of 28 rats, divided in control group (K), treatment group 1 (P1), treatment group 2 (P2), treatment group 3 (P3). All rats were adapted for 7 days, then on the 7th day we took the rat peripheral blood from retroorbital vein and calculate platelets count by Barbara Brown methods. Nigella sativa extracts were given on the 8th until 21st day, the doses are 2x0,008ml each day (P1), 2x0,08ml each day (P2), and 2x0,8ml each day (P3). All groups were given 0,54mg methotrexate each day on the 16th until 20th day. On the 22nd day we took the rat peripheral blood from retroorbital vein and calculate platelets count by Barbara Brown methods. Results: Platelets count of P2 group after treatment was decreased but it was not significant compared with the reduction of platelets count on K and P1. There was significantly different between P1 and P2 (p=0,028). Conclusion: Nigella sativa extract 2x0,08ml each day prevent the reduction of platelets count on Wistar rats given 0,54mg methothexate.
