Abstract
Background: Colon carcinogenesis induction by 1,2 DMH, high lipid and protein diet in Wistar rats is assumed having influence to their immunity system, causing injury on their spleen. Cellulose, being known as chemo-preventive agent for colon cancer, can reduce injury reaction on the spleen, because of the carcinogenesis process. The purpose of this research was to describe the effects of colon carcinogenesis induction, carcinogenesis induction plus cellulose dietary to Wistar’s spleen histopathological pattern. Methods: This laboratory observational study used 15 Wistars which divided into 3 groups. Each group consists of 5 Wistars with no intervention ( normal condition), 5 Wistars induced colon carcinogenesis, and 5 Wistars induced colon carcinogenesis and given dietary cellulose orally before and during the induction. At the ninth week, all rats were terminated, and samples of histopathological pattern were made. Data measured were macroscopic ( spleen’s volume and weigh ) and microscopic ( mean diameter of centrum germinativum, white pulp, and marginal zone and the amount of lymphocytes , plasma cells, and macrophages ). Data were analyzed descriptively then presented in table. and graphics according to their intervention. Results: The average weight of spleen for normal group is 0,33 gr, the volume is 0,38 ml, diameter of centrum germinativum 16µ , white pulp 31,6µ, and the distance of marginal zone is 1,4µ, the count of lymphocyte 29, plasma cell 0, and macrophage 1. For induction group, the average weight is 0,76gr, the volume 0,62ml, diameter of centrum germinativum is 12,38µ; white pulp 27,88µ; and the distance of marginal zone is 4,38µ, lymphocyte 23, plasma cell1 and macrophage 2; while for the cellulose group, the average weight is 0,52gr, the volume 0,65ml, diameter of centrum germinativum 13,67µ; white pulp 28,5µ;and the distance of marginal zone is 2,17µ, lymphocyte 33, plasma cell 1, macrophage 1. Conclusion: This research have described about the effects of; carcinogenesis induction, carcinogenesis induction plus cellulose dietary to Wistar’s spleen weight, volume, and histopathological pattern ( average diameter of white pulp, centrum germinativum, and distance of marginal zone, the count of lymphocyte, plasma cells and macrophage) and in normal condition / without intervention.
