Abstract

Background: Mahkota dewa (Phaleria macrocarpa) has been commonly used as a traditional herbal treatment for many diseases. Flavonoid, one of its main contents, is reported to have an immunostimulatory effect against intracellular pathogenic bacterial infections, such as Salmonella typhimurium. This effect showed by the increase of phagocytotic action. The activated macrophage would produce ROI to kill the phagocyted bacteria. Objective: To find out Phaleria macrocarpa could increase ROI production of macrophage in Balb/c mice infected by Salmonella typhimurium. Methods: An experimental laboratory study with a post test only-control group design was done. Samples were 30 female Balb/c mice, 6-8 weeks old, 20-25 grams, and without any physical abnormalities. Random alocation by simple random sampling into 6 groups: (K) was only given daily consumption. (P1) was given Phaleria macrocarpa 0.12 ml/day per oral. (P2) was infected by Salmonella typhimurium 106/0.1 cc at day-8. (P3), (P4), (P5) each was given Phaleria macrocarpa 0.06; 0.12; 0.24 ml/day per oral then infected by Salmonella typhimurium 106/0.1 cc. The ROI level was measured at day-13 using the NBT reduction method. Result: Mean of ROI level for (K)=0.337; (P1)=0.781; (P2)=0.597; (P3)=0.814; (P4)=1.194; (P5)=1.698. There was a significant increase showed by non parametric test in experimental group (p<0.05). Conclusion: Phaleria macrocarpa could significantly increase the ROI level of macrophage.
