Abstract

Background: Salmonella is the main cause of thypoid fever. Lymphocyte plays a major rule in specific immunity against Salmonella. Hedyotis corymbosa contains some active components such as flavonoid which can increase the production of IL-2, and as a result, increasing the lymphocyte proliferation. The maturation of lymphocyte takes place in the spleen, a secondary lymphoid organ. The objective of this research is to know the effect of Hedyotis corymbosa’s extract on spleen’s lymphocyte proliferation of Balb/c mice infected by Salmonella typhimurium. Method: A laboratory experimental study with post-test only control group design was carried out on experiment animal 30 male Balb/c mice which were randomly chosen. These mice divided into 6 groups. K1 was control group which is only given aquadest, K2 was control group infected with Salmonella typhimurium on the tenth day, whereas K3 was control group treated with Hedyotis corymbosa 80mg/day. P1, P2 and P3 were group given Hedyotis corymbosa 24mg, 80mg, and 240mg/day for 14 days then infected by Salmonella typhimurium on the tenth day. All mices were killed in day-15, and then lymphocytes from the spleen of all mices were isolated. Spleen’s lymphocyte proliferation was measured by counting lymphoblasts in every 200 cells in a homogenous area. Results: Significant differences are shown in lymphoblast count between K1-K2 (p=0.009), K1-P1 (p=0.018), K1-P2 (p=0.012), K1-P3 (p=0.016). There’s no significant differences in lymphoblast count between K3-K1 (p=0.059). There’s also no significant differences neither between P1 and P2 (p=0.745), P1 and P3 (p=0.289), nor between P2 and P3 (p=0.401) Conclusions: Hedyotis corymbosa’s extract cannot increase the lymphocyte proliferation on Balb/c mice infected by Salmonella typhimurium.
