Abstract

Background : Mahkota dewa contains chemical substances like flavanoid. Study had proven that flavanoid can increase IL-2 production that induces lymphocytes proliferation. Spleen is the largest lymphoid organ that produces lymphocytes. The aim of this study is to prove the effect of mahkota dewa extract on the increasing of spleen lymphocytes proliferation of BALB/C mice. Method : An experimental study with the post-test only control group design was carried out on 25 male BALB/C mice, which was divided into 5 groups. K was control group, whereas the treated groups were P1 which was given mahkota dewa extract 35 μg/day, P2 given 70 μg/day, P3 given 140 μg/day, and P4 given 280 μg/day. Lymphocytes from the spleen of all mice were isolated after 2 weeks treatment, then lymphoblasts were counted in every 200 cells. Results : There were significant differences in lymphoblasts count between K and P1 (p=0,009 and P2 (p=0,028); P1 and P2 (p=0,009), P3 (p=0,009), and P4 (p=0,009); P2 and P3 (p=0,012) and P4 (p=0,028). It may be caused by flavanoid. There were no significant differences in lymphoblasts count between K and P3 (p=0,917), K and P4 (p=0,675), P3 and P4 (p=0,917). It may be caused by the other effects of flavanoid that are cytotoxic and immunosuppressive, especially on high dose. Conclusion: On the giving of mahkota dewa extract there were significant increase of lymphocytes proliferation on P1 and P2 groups. The highest proliferation was shown on P1 group given 35 μg extract/day. Lower proliferation were shown on the groups given higher dose of extract.
