Abstract

Pengaruh Pemberian Suplemen Melatonin terhadap Jumlah Sel Busa dan Ketebalan Dinding Aorta Abdominalis Tikus Wistar yang Diinduksi Aterosklerosis Nugroho, Adimas Okto ; Hardian Latar Belakang : Penelitian terdahulu telah membuktikan bahwa hormon melatonin yang diproduksi oleh kelenjar pineal memiliki sifat antioksidan yang dapat menangkal radikal bebas. Radikal bebas merupakan salah satu faktor pemicu terjadinya proses aterogenesis. Pemberian diit kuning telor intermitten dan injeksi inisial adrenalin pada tikus Wistar terbukti dapat menimbulkan proses aterosklerosis. Penelitian ini bertujuan mengetahui pengaruh pemberian suplemen melatonin terhadap jumlah sel busa dan ketebalan dinding aorta abdominalis. Metode : Penelitian ini adalah penelitian aksperimental dengan rancangan Randomized Post-test Only Control Design. Penelitian ini menggunakan 16 tikus Wistar jantan, dibagi secara acak manjadi dua kelompok. Kelompok I ( perlakuan ) diberi injeksi inisial adrenalin bitatras sebanyak 0,006 mg i.v pada hari pertama, dilanjutkan dengan diet intermitten kuning telur 10 mg tiap dua hari sekali serta diet standar setiap hari dari hari ke-2 sampai hari ke-29, di hari ke-16 ditambah pemberian suplemen melatonin 0,0054 mg setiap hari sampai hari ke-29. Kelompok II (kontrol) diberi injeksi inisial adrenalin 0,006 mg i.v pada hari pertama, dilanjutkan dengan diet intermitten kuning telur 10 mg tiap dua hari sekali serta diet standar setiap hari dari hari ke-2 sampai hari ke-29 tanpa ada pemberian suplemen melatonin. Pada hari ke-30 tikus didekapitasi, diambil aorta abdominalis. Dilakukan pengecatan hematoksilin eosin, selanjutnya menghitung jumlah sel busa dan mengukur ketebalan dinding aorta abdominalis. Data dianalisa dengan uji Mann-Whitney untuk membedakan antar dua kelompok. Hasil : Rata-rata jumlah sel busa tikus Wistar kelompok I ( perlakuan ) yaitu 19,4 sel (SD = 7,17) lebih rendah dibandingkan kelompok II ( kontrol ) yaitu 28 sel (SD = 7,73). Rata-rata ketebalan dinding aorta abdominalis tikus Wistar kelompok I ( perlakuan ) yaitu 141,1 µ (SD = 31,18) lebih rendah dibandingkan kelompok II ( kontrol ) yaitu 184,7 µ (SD = 20,29). Uji Mann-Whitney data jumlah sel busa antar dua kelompok berbeda bermakna ( p=0,05 ), sedangkan uji Mann-Whitney data ketebalan dinding aorta antar dua kelompoknya berbeda bermakna ( p=0,015 ). Kesimpulan : Pemberian suplemen Melatonin dengan induksi aterosklerosis dapat menurunkan jumlah sel busa secara bermakna serta dapat menurunkan ketebalan dinding Aorta Abdominalis tikus Wistar secara bermakna. The Effect of Melatonin Supplement on Foam Cell Formation and The Abdominal Aortic Wall Thickness in Wistar with Atherosclerosis Induction Nugroho, Adimas Okto ; Hardian Background : The prior research has proved that Melatonin which produced by pineal gland contain antioxidant. Free radical is one of the primary causes of atherogenesis process. The intermitten egg yolk dietary and initial adrenalin injection in wistar were proved to increased a risk of atherosclerosis process. The aim of study was to prove the effect of melatonin supplement to the number of foam cell and the thickness of aortic wall. Method : The Randomized Post-test Only Control Design was chosen as a research design. The object of study were 16 wistars, devided into two group. Group I induced atherogenesis with initial injecion by 0,006 mg adrenalin on the first day, followed by standard diet/days and egg yolk dietary/2 days on the 2nd to 29th days, on the 16th to 29th days were given 0,0054 mg/days melatonin supplement. Group II induced atherogenesis with initial injecion by 0,006 mg adrenalin on the first day, followed by standard diet/days and egg yolk dietary/2 days on the 2nd to 29th days without melatonin therapy. On the 30th day they were decapited and their abdominal aortic were processed using hematoxylin eosin stained to examined the foam cell and the abdominal aortic thickness. The data were analyzed by Mann-whitney test to differences between group. Result : The study showed that the mean of foam cell in group I : 19,4 cell (SD = 7,17) were less than group II : 28 cell (SD = 7,73). The mean of abdominal aortic wall thickness in group I : 141,1 µ (SD = 31,18) were less than group II : 184,7 µ (SD = 20,29). Mann-Whitney test of the foam cell between group I and group II showed a significant difference (p=0,05). Mann-Whitney test of the abdominal aortic thickness between group I and group II showed a significant difference (p=0,015). Conclusion : The atherogenesis induction, followed by Melatonin supplement dietary was significantly decreased the number of foam cell and significantly decreased the thickness of aortic wall.
