Abstract
ANTHELMINTIC POTENCY TEST OF GARLIC (Allium sativum) TUBER SQUEEZE TO Ascaridia galli WORM IN VITRO Yenny Yulianti*, Noor Wijayahadi** Background: Allium sativum or garlic is a medical plant widely used in traditional medicine for the treatment of hypertension, diabetes, cough, itch, fungal infections, and worm infections. Garlic contains alisin, active subtance that consists of dialil disulfide, dialil trisulfide, propil alil disulfide, dialil mono sulfide, alil polisulfide, and squiterpene, with the existing substance, garlic is hoped to have an anthelmintic effect. In this research, the anthelmintic effect of garlic tuber squeeze to Ascaridia galli in vitro is compared using piperazine citrate solution as positive control and NaCl 0,9% solution as negative control. Method: This research was an experimental research with post test only control group design. The samples were 234 Ascaridia galli worms, which were divided into 3 groups. The first group was squeeze of garlic tuber with 10%, 25%, 50%, 60%, 75%, and 100% concentrations. The second group was piperazine citrate solutions in 0,2%, 0,3%, 0,4%, 0,5%, 0,6%, and 0,7% concentrations as positive control. The third group was NaCl 0,9% solutions as negative control. Each group was triple replicated. The volume of sample administered was 25 ml for each petri dish containing 6 worms. Each petri dish was incubated at 37oC, and then observed and recorded every 15 minutes for the total dead and or paralyzed worms. LC50 and LT50 of garlic tuber squeeze as anthelmintic was calculated using probit analysis methode. Treatment and control group data were analyzed by the test for differences, using SPSS 13.0 for Window, with significant level p<0,05. Result : Probit analysis showed that LC50 and LT50 of garlic (Allium sativum) tuber squeeze were 33,6477 % and 1 hour 38 minutes 0,2 seconds. Mann-Whitney test showed that treatment group and positive control group had significant difference (p<0,05) to negative control group. Garlic tuber squeeze 10% hadn’t significant difference (p>0,05) to positive control group. Garlic tuber squeeze 25%,50%.60%,75%,100% had significant difference (p<0,05) to positive control group. Conclusion: Garlic ( Allium sativum) tuber squeeze has in vitro anthelmintic effect to Ascaridia galli worm. Key Words: Anthelmintic, Allium sativum, Ascaridia galli. UJI DAYA ANTHELMINTIKPERASAN UMBI BAWANG PUTIH (Allium sativum)TERHADAP CACING Ascaridia galliSECARA IN VITRO Yenny Yulianti*, Noor Wijayahadi** Latar Belakang: Allium sativum (bawang putih) adalah tanaman obat yang secara luas digunakan dalam pengobatan tradisional untuk hipertensi, diabetes, batuk, gatal-gatal, infeksi jamur, dan infeksi cacing. Bawang putih mengandung alisin, bahan aktif yang terdiri dari dialil disulfida, dialil trisulfida, propil alil disulfida, dialil mono sulfida, alil polisulfida, dan squiterpene, dengan adanya zat tersebut diharapkan bawang putih mempunyai efek anthelmintik. Dalam penelitian ini, efek anthelmintik perasan umbi bawang putih terhadap cacing Ascaridia galli secara in vitro dibandingkan dengan larutan piperazine sitrat sebagai kontrol positif dan larutan NaCl 0,9% sebagai kontrol negatif. Metode: Penelitian ini merupakan penelitian eksperimental dengan post test only control group design. Sampelnya adalah 234 cacing Ascaridia galli, yang dibagi menjadi 3 kelompok. Kelompok pertama adalah perasan umbi bawang putih dengan konsentrasi 10%, 25%, 50%, 60%, 75%, dan 100%. Kelompok kedua adalah larutan piperazine sitrat dalam konsentrasi 0,2%, 0,3%, 0,4%, 0,5%, 0,6%, dan 0,7% sebagai kontrol positif. Kelompok ketiga adalah larutan NaCl 0,9% sebagai kontrol negatif. Tiap kelompok direplikasi 3 kali. Volume yang diberikan adalah 25 ml untuk tiap cawan petri yang berisi 6 ekor cacing. Setiap cawan petri diinkubasi pada suhu 37oC dan kemudian diamati dan dicatat setiap 15 menit jumlah cacing yang mati dan atau paralisis. LC50 dan LT50 perasan umbi bawang putih dihitung menggunakan metode analisis probit. Data kelompok perlakuan dan kelompok kontrol dianalisis dengan uji beda, menggunakan SPSS 13.0 for Windows, dengan taraf signifikasi p<0,05. Hasil: Dari hasil analisis probit diperoleh harga LC50 dan LT50 perasan umbi bawang putih (Allium sativum) adalah 33,6477 % dan 1 jam 38 menit 0,2 detik. Hasil uji Mann-Whitney menunjukkan bahwa kelompok perlakuan dan kelompok kontrol positif mempunyai perbedaan yang bermakna (p<0,05) terhadap kelompok kontrol negatif. Perasan umbi bawang putih 10% tidak mempunyai perbedaan yang bermakna (p>0,05) terhadap kelompok kontrol positif.Perasan umbi bawang putih 25%,50%,60%,75%,100% mempunyai perbedaan yang bermakna (p<0,05) terhadap kelompok kontrol positif. Kesimpulan: Perasan umbi bawang putih (Allium sativum) mempunyai daya anthelmintik terhadap cacing Ascaridia galli secara in vitro. Seiring dengan kenaikan konsentrasi perasan umbi bawang putih, maka semakin besar pula daya anthelmintiknya. Kata Kunci: Anthelmintik, Allium sativum, Ascaridia galli.
