Abstract

The Toxicity of Ganoderma lucidum Extract to Liver by Observing Bilirubin Rate in The Serum of Wistar Rat Rigina Nilandrania), M. Masjhoerb) Background: Former researches result in fat degeneracy in Wistar rat’s liver after giving of Ganoderma lucidum extract in 3 months. This research aimed to prove that giving of extract G. lucidum by subchronic research could raise bilirubin rate in Wistar rat’s serum and to find the cause by using fenobarbital as liver metabolism inducer and cimetidine as liver metabolism inhibitor. Methods: This research was an experimental study with the post test only control group design. The samples consist of 40 male Wistar rats which randomly divided into four groups: the first group (K) was given standard food, the second group (P1) was given G. lucidum extract, the third group (P2 )was given G. lucidum extract and fenobarbital, the fourth group (P3 ) was given G. lucidum extract and cimetidine. The doses of G. lucidum extract, fenobarbital, and cimetidine were 2,97 mg; 1,35 mg; 2,7 mg /rat/day. By the end of the fourth and the eighth week, retroorbital blood samples were taken from five rats in each group. Then the blood samples were sent to the laboratory for the inspection of conjugated, unconjugated, and total bilirubin serum rate. Results: The rate of total and conjugated bilitubin in all groups were elevated significantly (p<0,01). The highest rate of total and unconjugated bilirubin is P1: 11,56 (SD 0,31) and 8,11 (SD 0,30), followed by P2, then P3. Conclusions: Giving G. lucidum extract continuesly in two months is altered the rate of total and conjugated bilirubin. This elevation is more caused by the form substance then the metabolit of G. lucidum. Keywords: Ganoderma lucidum, bilirubin, toxicity. Masjhoerb) Latar Belakang: Hasil penelitian terdahulu didapatkan gambaran degenerasi lemak pada hepar tikus Wistar yang diberi ekstrak G. lucidum selama 3 bulan. Penelitian ini bertujuan untuk membuktikan bahwa pemberian ekstrak G. lucidum secara subkronis dapat meningkatkan kadar bilirubin serum tikus Wistar dan untuk mengetahui penyebab peningkatannya digunakan obat antara fenobarbital sebagai induser metabolisme hepar dan cimetidine sebagai inhibitor metabolisme hepar. Metode: Penelitian ini menggunakan rancangan experimental post test only control grup design. Sampel terdiri dari 40 ekor tikus Wistar, secara random dibagi menjadi empat kelompok: kelompok K diberi pakan standar, kelompok P1 diberi ekstrak G. lucidum, kelompok P2 diberi ekstrak G. lucidum dan fenobarbital, kelompok P3 diberi ekstrak G. lucidum dan cimetidine. Dosis ekstrak G. lucidum, fenobarbital, dan cimetidine yang dipakai adalah 2,97 mg; 1,35 mg; 2,7 mg /ekor/hari. Pada akhir minggu keempat dan minggu kedelapan, diambil sampel darah retroorbitalis dari lima ekor tikus per kelompok. Darah dikirim ke laboratorium untuk pemeriksaan kadar bilirubin total, terkonjugasi, dan tak terkonjugasi. Hasil: Terjadi peningkatan kadar bilirubin total dan terkonjugasi pada seluruh kelompok perlakuan (p<0,01). Peningkatan kadar bilirubin total dan terkonjugasi tertinggi terdapat pada kelompok P1: 11,56 (SD 0,31) dan 8,11 (SD 0,30), diikuti kelompok P2 lalu P3. Kesimpulan: Pemberian ekstrak G. lucidum meningkatkan kadar bilirubin total dan terkonjugasi. Peningkatan ini lebih disebabkan oleh substansi asal dibandingkan dengan metabolit aktif G. lucidum.
