Abstract

The Effect of Administering Lycopersicum esculentum Mill on Spermatozoa Viability of Male Balb/C Mice Exposed by Cigarette Smoke Hesti Dyah Palupi , Juwono Background : One of the cause of infertility is the excessive amount of free radicals. Free radicals create reactive ocxygen result in the abnormality of membrane and sperm motility. Lycopersicum esculentum Mill mainly consists of lycopen, flavonoid and vitamin C which work as antioxydant in body. Antioxydant is able to resist free radicals so it reduce abnormality spermatozoa. The objective of this study is to prove if antioxcydants in tomato can influence the spermatozoa viability male Balb/C mice exposed by cigarette smoke. Method : An experimental study using the post test only control group design. The sample were 36 male Balb/C mice with specific criteria. Sample were randomized into four groups (group O1, O2, O3 and O4) and last three group was given smoke per 30s/day. Group O1 as negative control without any treatment. Group O2 as positive control, group O3 was given Lycopersicum esculentum Mill with a dose of 1,7 gr/day and Group O4 with higher dose, 3,5 gr/day. On the 54th day, the mice were decapitated, then sperm taken from vas deferen were made into slides with Giemsa stain. This data were analyzed by One Way Anova. Result : The mean of spermatozoa viability of negative Control group O1 was 91,78±3,40, positive Control O2 was 76,18±8,56, Treatment group O3 was 92,35±6,18 and Treatment group O4 was 88,09±3,93. It was shown in this study that the percentage of spermatozoa viability of the treaten group was significantly higher than control (p<0,05). Conclusion : The administering of Lycopersicum esculentum Mill exposed with smoked cigarette in 53 days gives better result for spermatozoa viability of Balb/C mice by the dose of 1,7 gr/day and 3,5 gr/day. Keyword : Spermatozoa, tomato, cigarette smoke Pengaruh Pemberian Jus Buah Tomat Lycopersicum esculentum Mill Terhadap Viabilitas Spermatozoa Mencit Balb/C Jantan Yang Diberi Paparan Asap Rokok Hesti Dyah Palupi , Juwono Latar Belakang : Salah satu penyebab menurunnya kualitas sperma adalah radikal bebas berlebih. Radikal bebas membentuk oksigen reaktif yang menyebabkan kelainan pada membran dan motilitas spermatozoa. Lycopersicum esculentum Mill, dengan kandungan utama likopen, flavonoid dan vitamin C, bekerja sebagai antioksidan utama. Antioksidan menetralisir radikal bebas sehingga dapat mencegah kerusakan spermatozoa. Penelitian ini bertujuan untuk membuktikan pengaruh antioksidan dalam tomat terhadap viabilitas spermatozoa mencit Balb/C jantan yang diberi paparan asap rokok. Metode Penelitian : Penelitian eksperimental dengan post test only control group design. Jumlah sampel 36 ekor mencit Balb/C jantan dengan kriteria tertentu. Sample dirandom menjadi empat kelompok (Kontrol(-), Kontrol(+), Perlakuan O3 dan Perlakuan O4) dan 3 kelompok terakhir dipapar asap rokok selama 30 detik setiap hari. Kelompok O3 diberi Lycopersicum esculentum Mill dengan dosis 1,7 gr/hari dan kelompok O4 dengan dosis 3,5 gr/hari. Pada hari ke-54, mencit didekapitasi untuk diambil vas deferennya dan dibuat preparat dengan pengecatan Giemsa. Data diuji dengan One Way Anova. Hasil : Dari pembacaan preparat didapatkan rerata kelompok Kontrol(-) 91,27±3,40, Kontrol(+) 76,18±8,56, Perlakuan O3 sebesar 92,35±6,18 dan Perlakuan O4 88,09±3,93. Hasil uji ANOVA menunjukkan viabilitas spermatozoa kelompok Perlakuan memiliki peningkatan persentase secara signifikan (p<0,05) dari Kontrol (+). Kesimpulan : Pemberian Lycopersicum esculentum Mill pada mencit Balb/C dengan dosis 1,7 gr/hari dan dosis 3,5 gr/hari selama 53 hari dan dipapar asap rokok memberikan hasil persentase viabilitas spermatozoa yang lebih baik dibandingkan kelompok Kontrol (+). Dosis tomat 1,7 gr/hari memberikan hasil lebih baik dibandingkan dengan dosis 3,5 gr/hari pada mencit Balb/C jantan yang diberi paparan asap rokok. Kata Kunci : Spermatozoa, tomat, asap rokok
