PENGARUH PEMBERIAN JUS BUAH TOMAT (Lycopersicum esculentum Mill) TERHADAP MOTILITAS SPERMATOZOA MENCIT Balb/c JANTAN YANG DIBERI PAPARAN ASAP ROKOK

Abstract

The Effect of Tomato (Lycopersicum esculentum Mill) Juice on Spermatozoa Motility of Male Balb/c Mice Exposed by Cigarette Smoke Diana Imam*, Juwono** Background : An excess free radical is one of causes male infertility case. This radical can disturb human spermatozoa such as the abnormality of membrane and sperm motility. Antioxydant can netralize this free radical. Tomato (Lycopersicum esculentum Mill) is a fruit which has substance with highest antioxydants.The purpose of this study is to prove whether tomato juice (Lycopersicum esculentum Mill) can prevent the decreasing of the sperm motility of male Balb/c mice with smoke treatment. Methods:This experimental research used the post test only control group design. The sampel were 36 Balb/c male mice, which divided into four groups(K(+), K(-), O1, O2). Each of group was given meal standart. Control negative group was only given meal standart, Control positive(K) group was not given tomato juice but exposed by cigarette smoke, group O1 was given tomato juice with 1,7 gr/day and group O2 with dose 3,5 mg/day.This study lasted for 53 days. On the 54thall mice were sacrificed and the sperm motility was examined. Result :Mean of total motility sperm of three groups were group:K(-)=34,71; K(+)= 11,84 ;group O1= 49,24 ;group O2= 27,02.Oneway ANOVA test shown that there was decreasing significant difference between control positive group and control negative group (p<0,05). There was significant increasing difference between group K(+) and treatment group (p=0,00). Conclusion : Tomato juice (Lycopersicum esculentum mill) supplementation in 1,7 gr/day and 3,5 gr/day can prevent the decreasing of the sperm motility of male Balb/c mice exposed by cigarette smoke and the group with dose of 1,7 gr/day gives better result than 3,5 gr/day. Senyawa ini dapat memberikan gangguan pada spermatozoa antara lain: kelainan membran dan motilitas sperma. Senyawa antioksidan dapat menetralisir radikal bebas ini. Tomat (Lycopersicum esculentum Mill)merupakan salah satu buah dengan kandungan antioksidan tertinggi.Tujuan dari penelitian ini adalah membuktikan apakah jus buah tomat dapat mencegah penurunan motilitas spermatozoa mencit Balb/c jantan yang diberi paparan asap rokok. Metode :Penelitian ini menggunakan the post test only control group design. Jumlah sampel sebanyak 36 ekor mencit Balb/c jantan yang dibagi menjadi empat kelompok ( K(-),K(+),O1,O2). Tiap kelompok diberikan pakan standar. Kelompok K(-)merupakan kelompok tanpa perlakuan, kelompok K(+) hanya diberi paparan asap rokok, kelompok O1 diberi paparan asap rokok dan diberi jus buah tomat dengan dosis 1,7 gr/hari dan kelompok 02 juga diberi paparan asap rokok dan diberi paparan asap rokok dengan dosis 3,5 gr/hari.Perlakuan berlangsung selama 53 hari. Pada hari ke-54, semua mencit dibunuh lalu diperiksa motilitas spermatozoanya. Hasil :Rerata motilitas spermatozoa adalah sebagai berikut: Kelompok K(-)=34,71; kelompok K(+)=11,84; Kelompok O1= 49,24; Kelompok O2= 27,02. Uji Oneway ANOVA menunjukkan bahwa terdapat penurunan motilitas spermatozoa antara kelompok K(+) dengan kelompok K(-) yang signifikan (p<0,05) dan terdapat peningkatan motilitas spermatozoa antara kelompok K(+) dengan kelompok perlakuan yang signifikan (p<0,05) serta dosis 1,7 gr/hari memberikan hasil yang lebih baik dibandingkan dengan 3,5 gr/hari pada mencit Balb/c jantan yang dipapar asap rokok. Kesimpulan :Pemberian jus tomat (Lycopersicum esculentum Mill) dapat mencegah penurunan motilitas spermatozoa mencit Balb/c jantan yang diberi paparan asap rokok dan dosis jus tomat 1,7 mg/hari memberikan hasil yang lebih baik dibandingkan dengan dosis 3,5 mg/hari pada mencit Balb/c jantan yang diberi paparan asap rokok.
